【摘要】：目的建立HPLC-MS/MS法測定人血漿中氫嗎啡酮的質(zhì)量濃度。方法用固相萃取法處理血漿,用Kromasil 100-5SIL和E97915色譜柱,以95%乙腈-5 mmol·L-1乙酸銨為流動相,梯度洗脫,用正離子、多反應(yīng)監(jiān)測方式測定樣品質(zhì)量濃度。結(jié)果血漿樣品中,氫嗎啡酮在0.05~10.00 ng·m L-1內(nèi)線性關(guān)系良好(r=0.999 8),最低定量下限為0.05 ng·m L-1。血樣日內(nèi)與日間RSD均小于15%,平均回收率70%,且穩(wěn)定性均較好。結(jié)論本方法簡便快速、靈敏準(zhǔn)確、特異性強(qiáng),適用于氫嗎啡酮的體內(nèi)藥代動力學(xué)研究。
[Abstract]:Objective to establish a HPLC-MS/MS method for the determination of hydromorphone in human plasma.Methods the plasma was treated by solid phase extraction. The plasma was treated with Kromasil 100-5SIL and E97915 columns. The mobile phase was 95% acetonitrile-5 mmol L-1 ammonium acetate, gradient elution was carried out, and the mass concentration of the sample was determined by positive ion and multi-reaction monitoring method.Results in plasma samples, the linear range of hydromorphine was 0.05 ~ 10.00 ng mL ~ (-1) and the lowest limit of determination was 0.05 ng / mL ~ (-1).The average recovery rate was 70 and the stability was good.Conclusion the method is simple, rapid, sensitive, accurate and specific. It is suitable for the in vivo pharmacokinetics of hydromorphine.
