基于PC12細(xì)胞模型分析大豆蛋白水解物對神經(jīng)元氧化損傷的保護(hù)作用

【摘要】:本文基于PC12細(xì)胞模型研究大豆蛋白水解物(Soybean protein isolate hydrolysates,SPIHs)對神經(jīng)元氧化損傷的保護(hù)作用。以大豆蛋白為原料,經(jīng)過酶解和膜分離得到四種分子量不同的水解物,我們首先檢測了SPIHs的抗氧化能力;然后用H2O2刺激P12細(xì)胞,建立神經(jīng)元氧化損傷模型,并以適當(dāng)濃度的SPIHs處理細(xì)胞,通過檢測各種生物學(xué)指標(biāo)評價對細(xì)胞氧化損傷的保護(hù)作用。結(jié)果顯示,低分子量的SPIHs表現(xiàn)出最強(qiáng)的抗氧化活性;能夠提高損傷細(xì)胞的存活率和抗氧化酶活力,減少乳酸脫氫酶(LDH)的釋放量和丙二醛(MDA)的生成,抑制細(xì)胞活性氧(ROS)的累積(p0.05或p0.01),且變化呈現(xiàn)一定的劑量依賴關(guān)系。研究認(rèn)為,低分子量的SPIHs對神經(jīng)元氧化損傷具有保護(hù)作用,可以作為功能性成分用于保護(hù)神經(jīng)元氧化損傷相關(guān)的功能食品和保健品的開發(fā)。
[Abstract]:The protective effect of soybean protein hydrolysate (Soybean protein isolate hydrolysates,SPIHs on oxidative damage of neurons was studied based on PC12 cell model. Using soybean protein as raw material, four kinds of hydrolysates with different molecular weight were obtained by enzyme hydrolysis and membrane separation, and then the antioxidant capacity of SPIHs was measured, and then the oxidative damage model of neurons was established by stimulating P12 cells with H2O2, and the cells were treated with appropriate concentration of SPIHs to evaluate the protective effect on oxidative damage of P12 cells by testing various biological indexes. The results showed that low molecular weight SPIHs showed the strongest antioxidant activity, which could increase the survival rate and antioxidant enzyme activity of injured cells, reduce the release of lactic dehydrogenase (LDH) and the production of malondialdehyde (MDA), and inhibit the accumulation of reactive oxygen species (ROS) (p0.05 or p0.01) in a dose-dependent manner. It is considered that low molecular weight SPIHs has protective effect on neuronal oxidative damage and can be used as a functional component to protect the development of functional food and health products related to neuronal oxidative damage.
