鹽酸苯環(huán)壬酯對大鼠后循環(huán)缺血性眩暈和小鼠位置性眩暈的改善作用

【摘要】:目的研究鹽酸苯環(huán)壬酯的抗眩暈作用。方法利用雙側(cè)頸總動脈結(jié)扎法制備大鼠腦缺血模型,大鼠ig給予鹽酸苯環(huán)壬酯0.1~4.0 mg·kg~(-1),每天2次,連續(xù)3 d,結(jié)合激光多普勒腦血流儀檢測鹽酸苯環(huán)壬酯對腦血流量變化的影響。通過XY-5雙軸變速旋轉(zhuǎn)刺激制備小鼠眩暈癥模型,小鼠ig給予鹽酸苯環(huán)壬酯1.4~5.6 mg·kg~(-1)后變速旋轉(zhuǎn)刺激30 min,檢測自發(fā)活動的變化;小鼠ig給予鹽酸苯環(huán)壬酯1.4~8.4 mg·kg~(-1)30 min后,通過檢測胃酚紅排空率指標觀察鹽酸苯環(huán)壬酯對小鼠胃平滑肌的影響。結(jié)果大鼠雙側(cè)頸總動脈結(jié)扎24 h后腦血流量顯著減少(P0.01);與模型組比較,鹽酸苯環(huán)壬酯0.5,2.0和4.0 mg·kg~(-1)可顯著增加缺血大鼠腦血流量(P0.05)。通過旋轉(zhuǎn)刺激后,模型小鼠自發(fā)活動的總路程減少(P0.05),運動速度減慢(P0.05);與模型組比較,鹽酸苯環(huán)壬酯2.8和5.6 mg·kg~(-1)可增加眩暈小鼠總路程和運動速度(P0.05);鹽酸苯環(huán)壬酯5.6和8.4 mg·kg~(-1)降低小鼠胃酚紅排空率(P0.05)。結(jié)論鹽酸苯環(huán)壬酯增加腦血流量,改善后循環(huán)缺血,對眩暈的嘔吐癥狀和運動功能具有顯著的改善作用。
