二氫楊梅素對(duì)濕性年齡相關(guān)性黃斑變性的影響

【摘要】:目的:探討二氫楊梅素(DMY)對(duì)濕性年齡相關(guān)性黃斑變性(AMD)的影響。方法:用532 nm激光對(duì)成年日本大耳白兔眼底進(jìn)行光凝,創(chuàng)建AMD模型。光凝4周后,DMY(100 mg·kg~(-1)·d~(-1))灌胃給藥,酶聯(lián)免疫吸附法(ELISA)檢測(cè)外周血中血管內(nèi)皮生長(zhǎng)因子(VEGF)和色素上皮衍生因子(PEDF)水平;實(shí)驗(yàn)結(jié)束時(shí),蘇木精-伊紅染色法檢測(cè)脈絡(luò)膜新生血管(CNV);解剖實(shí)驗(yàn)動(dòng)物,分別計(jì)算肝臟、腎臟和脾臟指數(shù)。結(jié)果:給藥4周,模型組外周血VEGF降低幅度為63.3%,而模型對(duì)照組降低幅度為48.6%;給藥8周,模型組外周血VEGF降低幅度為46.7%,模型對(duì)照組降低幅度為20.1%。給藥4周,模型組外周血PEDF降低幅度為55.9%,而模型對(duì)照組降低幅度為58.6%;給藥8周,模型組外周血PEDF降低幅度為12.1%,模型對(duì)照組降低幅度為53.1%。給藥8周,模型對(duì)照組可見(jiàn)明顯CNV,而模型給藥組未見(jiàn)明顯CNV;模型給藥組肝臟指數(shù)和脾臟指數(shù)明顯低于模型對(duì)照組(P=0.147和P=0.223),模型給藥組腎臟指數(shù)與模型對(duì)照組沒(méi)有明顯差異(P=0.874)。結(jié)論:DMY具有一定的抑制濕性AMD模型日本大耳白兔CNV生長(zhǎng)的藥理作用,其作用機(jī)制可能與其下調(diào)VEGF的表達(dá)、上調(diào)PEDF表達(dá)和益氣活血、健脾疏肝有關(guān)。
[Abstract]:Objective: to investigate the effect of dihydromyricetin (DMY) on (AMD) of wet age related macular degeneration. Methods: the AMD model was established by photocoagulation on the fundus of adult Japanese white rabbits with 532 nm laser. After 4 weeks of photocoagulation, DMY (100 mg kg~ (-1) d ~ (-1) was administered intragastrically. The levels of vascular endothelial growth factor (VEGF) and pigment epithelium-derived factor (PEDF) in peripheral blood were detected by enzyme-linked immunosorbent assay (ELISA). At the end of the experiment, hematoxylin-eosin staining was used to detect the choroidal neovascularization (CNV); in experimental animals, and the indexes of liver, kidney and spleen were calculated. Results: after 4 weeks of administration, the level of VEGF in peripheral blood of model group was 63.3%, while that of model control group was 48.6%. After 8 weeks of administration, the decrease of VEGF in peripheral blood of model group was 46.7%, and that of model control group was 20.1%. After 4 weeks of administration, the decrease of PEDF in peripheral blood of model group was 55.9, that of model control group was 58.6, and that of peripheral blood PEDF was 12.1 in model group and 53.1 in model control group after 8 weeks of administration. After 8 weeks of administration, significant CNV, was found in the model control group, but no significant CNV; was found in the model administration group. The liver index and spleen index in the model group were significantly lower than those in the model control group (P < 0. 147 and P < 0. 223), but there was no significant difference between the model group and the model control group (P < 0. 874). Conclusion: DMY has a certain pharmacological effect on inhibiting the growth of CNV in Japanese large ear white rabbits with wet AMD model, and its mechanism may be related to its down-regulation of VEGF expression, up-regulation of PEDF expression, invigorating qi and activating blood circulation, and invigorating spleen and soothing liver.
