【摘要】：目的建立以紫外分光光度法檢測(cè)金屬β-內(nèi)酰胺酶活性的方法。方法采用以美羅培南作反應(yīng)底物,中性羥胺與硫酸鐵胺為顯色體系,通過紫外分光光度計(jì)檢測(cè)反應(yīng)液中美羅培南剩余量計(jì)算金屬β-內(nèi)酰胺酶的分解量,從而計(jì)算出其酶活性。結(jié)果美羅培南的濃度在1~5mg/m L范圍內(nèi),線性良好,相關(guān)系數(shù)大于0.999。該方法檢測(cè)限為0.05U,回收率在90%~119%范圍內(nèi),重復(fù)性相對(duì)標(biāo)準(zhǔn)偏差為6.1%~8.7%.檢測(cè)出的酶活性與酶蛋白含量成正相關(guān)。結(jié)論該方法能有效區(qū)分金屬β-內(nèi)酰胺酶與非金屬β-內(nèi)酰胺酶,并準(zhǔn)確反應(yīng)金屬β-內(nèi)酰胺酶分解碳青霉烯類抗生素的能力。
[Abstract]:Objective to establish a method for the determination of metal 尾-lactamases by ultraviolet spectrophotometry. Methods using meropenem as the substrate, neutral hydroxylamine and ferric sulfate as color reaction system, the decomposition amount of metal 尾 -lactamase was calculated by ultraviolet spectrophotometer to calculate the activity of 尾 -lactamase. Results the concentration of meropenem was linear in 1~5mg/m L range, and the correlation coefficient was greater than 0.999 9. The detection limit of this method is 0.05 U, the recovery is within 90% and the relative standard deviation of reproducibility is 6.1% and 8.7%. The activity of enzyme was positively correlated with the content of enzyme protein. Conclusion this method can effectively distinguish metal 尾 -lactamases from non-metallic 尾 -lactamases, and accurately react the ability of metal 尾 -lactamases to decompose carbapenem antibiotics.
