【摘要】:隨著全球癌癥患病比例的逐年增加,癌癥已經(jīng)成為全球死亡的主要原因和重大的公共衛(wèi)生問(wèn)題?拱╇哪芷茐哪[瘤細(xì)胞膜結(jié)構(gòu)或抑制癌細(xì)胞增殖和遷移以及腫瘤血管的形成,幾乎不表現(xiàn)溶血性且對(duì)正常的人體細(xì)胞基本無(wú)損傷等優(yōu)點(diǎn),已經(jīng)成為抗腫瘤新藥研究的一大熱點(diǎn)?拱╇闹饕獊(lái)源可分為4類(lèi):抗菌肽類(lèi)如牛乳鐵蛋白肽(Lfcin B)、天蠶素等,對(duì)細(xì)菌等微生物具有廣譜高效的抑制活性,同時(shí)能夠抑制多種癌細(xì)胞的增殖和生長(zhǎng),誘導(dǎo)癌細(xì)胞凋亡;多肽激素類(lèi)如心房利鈉肽(ANP)以及在ANP形成過(guò)程中的其他一些多肽,在臨床上具有增加血管通透性、降壓、利鈉、利尿、舒張血管平滑肌、調(diào)節(jié)水鹽平衡和抑制細(xì)胞增殖等作用,最近研究發(fā)現(xiàn)它們可以直接殺死腫瘤細(xì)胞或強(qiáng)力抑制癌細(xì)胞DNA的合成,從而抑制癌細(xì)胞的生長(zhǎng);多肽毒素類(lèi)如蜂毒、蛇毒、蝎毒等毒性較強(qiáng)的生物毒素,對(duì)多種腫瘤均有抑制作用;內(nèi)皮抑制素可以抑制腫瘤血管的生成抑制癌細(xì)胞的生長(zhǎng)和轉(zhuǎn)移?拱╇闹饕ㄟ^(guò)誘導(dǎo)細(xì)胞凋亡、破壞細(xì)胞的膜結(jié)構(gòu),改變細(xì)胞周?chē)蚣?xì)胞內(nèi)p H以及增強(qiáng)免疫應(yīng)答等多種機(jī)制來(lái)抑制癌細(xì)胞增殖。綜述抗癌肽的來(lái)源、抑癌機(jī)理,并對(duì)抗癌肽的應(yīng)用前景進(jìn)行分析,旨為抗癌肽的深入研究和開(kāi)發(fā)應(yīng)用提供參考。
[Abstract]:With the increasing incidence of cancer worldwide, cancer has become a major cause of death and a major public health problem. Anticancer peptides can destroy the structure of tumor cell membrane or inhibit the proliferation and migration of cancer cells, as well as the formation of tumor blood vessels, almost no hemolysis and no damage to normal human cells, which has become a hot spot in the research of new anti-tumor drugs. The main sources of anticancer peptides can be divided into four categories: antimicrobial peptides, such as lactoferrin Bu, daponin, have broad-spectrum and high efficiency inhibitory activity against bacteria and other microorganisms, and can inhibit the proliferation and growth of many kinds of cancer cells and induce apoptosis of cancer cells; Peptide hormones such as atrial natriuretic peptide (ANPP) and other peptides in the formation of ANP have the effects of increasing vascular permeability, lowering blood pressure, natriuretic, diuretic, relaxing vascular smooth muscle, regulating water and salt balance and inhibiting cell proliferation, etc. Recent studies have found that they can directly kill tumor cells or strongly inhibit the synthesis of DNA from cancer cells, thereby inhibiting the growth of cancer cells; polypeptide toxins such as bee venom, snake venom, scorpion venom and other highly toxic biotoxins, Endothelin inhibits the angiogenesis of tumor and inhibits the growth and metastasis of cancer cells. Anticancer peptides inhibit the proliferation of cancer cells by inducing cell apoptosis, destroying cell membrane structure, changing pH around or within cells and enhancing immune response. The origin and mechanism of anticancer peptides are reviewed, and the application prospect of anticancer peptides is analyzed in order to provide reference for further research and development of anticancer peptides.
