【摘要】：反向分子對接以小分子化合物(天然產(chǎn)物、先導(dǎo)化合物及化學(xué)合成物)為探針,在已知結(jié)構(gòu)的靶點(diǎn)數(shù)據(jù)庫內(nèi)搜尋可能與之結(jié)合的生物大分子,通過空間和能量匹配相互識別形成分子復(fù)合物,進(jìn)而預(yù)測藥物潛在的作用靶點(diǎn)。本研究運(yùn)用Discovery Studio3.5軟件的反向找靶(Target Fishing)功能模塊、結(jié)合蛋白質(zhì)數(shù)據(jù)庫(PDB database)對我們民族植物學(xué)創(chuàng)新團(tuán)隊(duì)所采集到的少數(shù)民族藥用植物地膽草(Elephantopus scaber L.)、毛大丁草(Gerbera piloselloides L.)、三葉鬼針草(Bidens pilosa L.)、閉花耳草(Hedyotis cryptantha Dunn)、升麻(Actaea cimicifuga L.)、青藤仔(Jasminum nervosum Lour.)中所提取的化學(xué)成分(包括一些新發(fā)現(xiàn)的化合物)進(jìn)行抗腫瘤和抗癌活性的驗(yàn)證,并利用分子對接技術(shù)考察化合物與受體之間的匹配情況。結(jié)果表明:以上植物中所提取的化合物與具有抗癌和抗腫瘤作用的藥效團(tuán)的匹配值都非常高,表明這些植物都具有較高的抗腫瘤及抗癌活性,為少數(shù)民族藥用植物在抗腫瘤和抗癌藥物的研發(fā)提供理論基礎(chǔ)。
[Abstract]:Reverse molecular docking uses small molecule compound ( natural product , lead compound and chemical composition ) as probe to search the biological macromolecules which may be bound to it in the target database of known structure . It can be used to identify the potential target of drug . The chemical constituents extracted from above plants ( including some newly discovered compounds ) are verified by anti - tumor and anti - cancer activity , and the matching between the compound and the receptor is investigated by means of molecular docking technology . The results show that the compounds extracted from the above plants have high matching values with the pharmacophore with anti - cancer and anti - tumor effects , suggesting that these plants have higher antitumor and anti - cancer activity , and provide theoretical basis for the research and development of anti - tumor and anti - cancer drugs for ethnic minority medicinal plants .
