【摘要】：目的:建立測定鹽酸多奈哌齊口腔崩解片中有關(guān)物質(zhì)的方法。方法:采用反相高效液相色譜(RP-HPLC)加校正因子的主成分自身對照法。色譜柱為Eternity-C18柱,流動相為3.85 mol/L癸烷磺酸鈉溶液-乙腈(65∶35,V/V)(高氯酸調(diào)p H=1.8),流速為1.4 ml/min,檢測波長為271 nm,柱溫為35℃,進(jìn)樣量為20μl。測定鹽酸多奈哌齊和雜質(zhì)Ⅰ、Ⅲ、Ⅳ、Ⅴ、Ⅵ、Ⅶ、Ⅷ的線性方程,以斜率計算雜質(zhì)相對于鹽酸多奈哌齊的校正因子,用相對保留時間確定各雜質(zhì)位置。結(jié)果:鹽酸多奈哌齊雜質(zhì)Ⅰ、Ⅲ、Ⅳ、Ⅴ、Ⅵ、Ⅶ、Ⅷ的相對保留時間分別為0.21、0.93、0.31、1.18、0.54、2.16、0.68,校正因子分別為0.51、0.93、0.87、1.17、1.05、1.31、1.39。測得3批供試品中雜質(zhì)Ⅷ的含量分別為0.041%、0.040%、0.043%,其他雜質(zhì)均低于檢測限;與雜質(zhì)對照品法測定結(jié)果一致。結(jié)論:該方法專屬性強、靈敏度高,可方便快速、準(zhǔn)確有效地測定鹽酸多奈哌齊口腔崩解片中有關(guān)物質(zhì)的含量。
