人臍帶間充質(zhì)干細(xì)胞生物學(xué)特性及脈沖電磁場(chǎng)促進(jìn)其成骨分化研究

【摘要】: 間充質(zhì)干細(xì)胞是一群中胚層來源的具有自我更新和多向分化潛能的多能干細(xì)胞。來自于胎兒期的間充質(zhì)干細(xì)胞具有更強(qiáng)大的增殖能力、更廣譜的分化潛能、更低的免疫原性等。有望作為組織工程或細(xì)胞替代治療優(yōu)良的種子細(xì)胞。 本實(shí)驗(yàn)從流產(chǎn)胎兒(胎齡12~18周)臍帶沃頓膠中分離人臍帶間充質(zhì)干細(xì)胞,對(duì)其形態(tài)學(xué)、細(xì)胞周期、生長(zhǎng)動(dòng)力學(xué)、免疫表型及多分化潛能(向脂肪細(xì)胞、軟骨細(xì)胞、成骨細(xì)胞等分化)等生物學(xué)特性進(jìn)行研究,證實(shí)所分離的細(xì)胞為間充質(zhì)干細(xì)胞。 以脈沖電磁場(chǎng)作為刺激hUCMSCs成骨分化的手段,比較脈沖電磁場(chǎng)方式和化學(xué)誘導(dǎo)方式對(duì)hUCMSCs分化為成骨細(xì)胞的速度、基因表達(dá)等方面的差異,明確PEMFs刺激hUCMSCs體外成骨分化能力;建立兔撓骨缺損模型,hUCMSCs與羥基磷灰石混合后注入兔撓骨缺損處,PEMFs每天刺激3h,觀察骨缺損再生情況,探討PEMFs促進(jìn)hUCMSCs體內(nèi)成骨分化能力。從而為骨組織工程和細(xì)胞替代治療提供新的種子細(xì)胞來源;證實(shí)PEMFs促進(jìn)hUCMSCs成骨分化的可行性。本實(shí)驗(yàn)研究結(jié)果為實(shí)現(xiàn)hUCMSCs的常規(guī)化分離培養(yǎng)提供了有價(jià)值的實(shí)驗(yàn)依據(jù);為驗(yàn)證hUCMSCs具有多分化潛能提供有力依據(jù);為證實(shí)PEMFs是促進(jìn)hUCMSCs成骨分化的有效手段提供了實(shí)驗(yàn)依據(jù);為PEMFS誘導(dǎo)hUCMSCs向成骨細(xì)胞分化的臨床應(yīng)用提供理論依據(jù)和技術(shù)支持。
[Abstract]:Mesenchymal stem cells are a group of mesoderm derived pluripotent stem cells with self-renewal and multi-differentiation potential. Mesenchymal stem cells from fetal stage have stronger proliferative ability, wider differentiation potential, lower immunogenicity and so on. It is expected to be a good seed cell for tissue engineering or cell replacement therapy. In this study, human umbilical cord mesenchymal stem cells were isolated from aborted fetus (12 ~ 18 weeks of gestational age) with Wharton gum. Morphology, cell cycle, growth kinetics, immunophenotype and multi-differentiation potential (adipocytes, chondrocytes) of human umbilical cord mesenchymal stem cells were studied. The biological characteristics of osteoblasts were studied, and the isolated cells were confirmed as mesenchymal stem cells. Using pulsed electromagnetic field as the means to stimulate the osteogenic differentiation of hUCMSCs, the difference of the speed and gene expression of pulsed electromagnetic field and chemical induction on the differentiation of hUCMSCs into osteoblasts in vitro was compared, and the ability of PEMFs to stimulate the osteogenic differentiation of hUCMSCs in vitro was determined. The rabbit model of scratching bone defect was established, and then injected with PEMFs into rabbit flexor bone defect for 3 hours every day. The regeneration of bone defect was observed, and the ability of PEMFs to promote osteogenic differentiation of hUCMSCs in vivo was investigated. These results provide a new source of seed cells for bone tissue engineering and cell replacement therapy, and confirm the feasibility of PEMFs in promoting osteogenic differentiation of hUCMSCs. The results of this study provide valuable experimental basis for the conventional isolation and culture of hUCMSCs, provide a powerful basis for verifying the multi-differentiation potential of hUCMSCs, and provide experimental basis for confirming that PEMFs are an effective means to promote the osteogenic differentiation of hUCMSCs. To provide theoretical basis and technical support for the clinical application of PEMFS induced differentiation of hUCMSCs into osteoblasts.
