種植體周圍炎對(duì)頜骨成骨細(xì)胞生物學(xué)功能的影響

【摘要】:目的研究種植體周圍炎條件下炎性微環(huán)境對(duì)頜骨成骨細(xì)胞生物學(xué)功能的影響。方法原代分離培養(yǎng)和純化種植體周圍炎來(lái)源和正常組織來(lái)源的人頜骨成骨細(xì)胞,取第3代細(xì)胞進(jìn)行鑒定和檢測(cè),MTT法檢測(cè)成骨細(xì)胞的增殖能力,實(shí)時(shí)定量聚合酶鏈反應(yīng)法檢測(cè)成骨細(xì)胞相關(guān)基因骨鈣素(OCN)、Runx2、Ⅰ型膠原(ColⅠ)的表達(dá),Western blot法檢測(cè)成骨細(xì)胞相關(guān)蛋白OCN的表達(dá)。結(jié)果從培養(yǎng)第4天起,種植體周圍炎來(lái)源的成骨細(xì)胞的增殖活力明顯低于正常組織來(lái)源的成骨細(xì)胞(P0.05)。培養(yǎng)7 d時(shí),與正常組織來(lái)源的成骨細(xì)胞相比,種植體周圍炎來(lái)源的成骨細(xì)胞相關(guān)基因OCN、Runx2、ColⅠ的表達(dá)均顯著降低(P0.05),相關(guān)蛋白OCN的表達(dá)也顯著降低(P0.05)。結(jié)論種植體周圍炎的炎性微環(huán)境降低了頜骨成骨細(xì)胞的增殖及分化能力。
