【摘要】：TORCH是一類引起新生兒出生缺陷的微生物的總稱,分別代表弓形蟲、風疹病毒、巨細胞病毒和單純皰疹病毒Ⅱ型和其他一些感染因素。這五種微生物在孕期感染婦女可以引起流產(chǎn)、早產(chǎn)及胎兒生長遲緩和發(fā)育畸形。由TORCH感染引起的孕期感染是一種最常見的先天性感染性疾病。雖然TORCH感染在懷孕早期會有一些輕微癥狀,但是如何在孕早期就發(fā)現(xiàn)并給予合理治療仍然是人們面臨的一個巨大挑戰(zhàn)。目前,在尚無有效疫苗和治療方法出現(xiàn)以前,對于孕婦TORCH感染的早發(fā)現(xiàn)、早診斷、早預防成為主要途徑。 本課題將這TORCH感染所涉及的五種微生物蛋白質(zhì)分子按預先設置的排列方式固定于載體表面形成微陣列,根據(jù)蛋白質(zhì)分子間特異性結(jié)合的原理,通過免疫膠體金構(gòu)建顯色系統(tǒng)和冷CCD成像模式,實現(xiàn)生物分子的簡單、準確、快速、大信息量的檢測。從microbix公司購回五種微生物的滅活抗原物質(zhì),用考馬斯亮藍G250法測蛋白的含量;用ELISA法檢測五種蛋白的效價;用吸收-抑制實驗檢測五種抗原的特異性。通過優(yōu)
[Abstract]:TORCH is a class of microbes that cause birth defects. They represent Toxoplasma, rubella virus, cytomegalovirus and herpes simplex virus type II and other factors. These five microbes can cause abortion, premature birth, fetal growth retardation and developmental malformation in pregnant women. The pregnancy induced by TORCH infection Infection is one of the most common congenital infection diseases. Although TORCH infection has some mild symptoms in the early pregnancy, it is still a great challenge to find and give reasonable treatment in the early pregnancy. The early discovery of TORCH infection in pregnant women before the emergence of effective vaccines and treatments has been found. Early diagnosis and early prophylaxis are the main ways.
In this project, five kinds of microbiological proteins involved in the TORCH infection are fixed on the surface of the carrier to form microarrays. According to the principle of specific binding between proteins, the color system and cold CCD imaging mode are constructed by immuno colloid gold, and the simple, accurate, rapid and large information amount of the biomolecules is realized. Testing the inactivated antigen of five kinds of microbes from microbix company, the content of protein was measured by Coomassie brilliant blue G250 method, the titer of five proteins was detected by ELISA method, and the specificity of five antigens was detected by absorption inhibition test.
