牛場肥水灌溉對土壤nirK、nirS型反硝化微生物群落結(jié)構(gòu)的影響

【摘要】:以徐水縣梁家營長期定位施肥試驗(yàn)田為研究對象,利用末端限制性片段長度多態(tài)性(T-RFLP)分析和克隆文庫構(gòu)建,研究了5種施肥處理(清水灌溉CK、無機(jī)肥灌溉CF、牛場肥水不同濃度、不同次數(shù)灌溉T4、T5和T11)下土壤中nirK、nirS型反硝化細(xì)菌群落多樣性及其群落結(jié)構(gòu)的演變。結(jié)果表明,不同施肥處理下nirK、nirS型反硝化細(xì)菌群落多樣性無顯著差異,但群落結(jié)構(gòu)卻有明顯變化:nirK型反硝化細(xì)菌群落結(jié)構(gòu)既受施肥種類又受施肥量影響,優(yōu)勢種群尤其對施肥種類和施肥量響應(yīng)顯著;nirS型反硝化細(xì)菌則主要受施肥種類影響,施肥量影響微弱。牛場肥水處理和無機(jī)肥處理分別促進(jìn)和抑制不同的nirS型反硝化細(xì)菌,群落主成分受無機(jī)肥促進(jìn)、牛場肥水抑制。系統(tǒng)發(fā)育分析結(jié)果表明,土壤中nirK型反硝化細(xì)菌主要與假單胞菌屬(Pseudomonas)、產(chǎn)堿桿菌屬(Alcaligenes)和根瘤菌屬(Rhizobium)的反硝化細(xì)菌具有較近的親緣關(guān)系;nirS型反硝化細(xì)菌主要與勞爾氏菌(Ralstonia)和紅長命菌屬(Rubrivivax)有較近的親緣關(guān)系。試驗(yàn)土壤中反硝化微生物多與目前已報(bào)道的好氧反硝化細(xì)菌親緣關(guān)系較近,這可能與微生物分析取自表層土有關(guān)。
[Abstract]:The long term location fertilization field of Xushui County was used to analyze and clone library by terminal restriction fragment length polymorphism (TRFLP). NirK in soil was studied under five fertilization treatments (irrigating CK with water, irrigating CFCs with inorganic fertilizer, irrigating with different concentrations of water in cattle farms, and irrigating T4N T5 and T11 with different times). The diversity of denitrifying bacteria community of nirS type and the evolution of its community structure. The results showed that there was no significant difference in diversity of denitrifying bacteria community of nirS type denitrifying bacteria under different fertilization treatments. However, the community structure of the denitrifying bacteria of the type of denitrifying bacteria was obviously changed, and the community structure of denitrifying bacteria was not only affected by the type of fertilization but also by the amount of fertilizer. The dominant population was especially responsive to the type of fertilization and the amount of fertilizer. NirS type denitrifying bacteria were mainly affected by the type of fertilization, but the effect of fertilizer amount was weak. Cattle farm fertilizer water treatment and inorganic fertilizer treatment promoted and inhibited different nirS denitrifying bacteria respectively. The results of phylogenetic analysis showed that the nirK type denitrifying bacteria mainly belonged to Pseudomonas spp. The denitrifying bacteria (Alcaligenes) and Rhizobium (Rhizobium) were closely related to each other. NirS denitrifying bacteria are mainly associated with Ralstonia and Rubrivivax. Most of the denitrifying microbes in the soil were closely related to the aerobic denitrifying bacteria reported so far. This may be related to microbial analysis taken from surface soil.
