【摘要】：為了挖掘可用于棉花花器官或纖維發(fā)育研究的候選基因,基于已發(fā)布的陸地棉全基因組數(shù)據(jù),利用生物信息學(xué)的分析方法鑒定了陸地棉WOX(WUSCHEL-related homeobox,WOX)轉(zhuǎn)錄因子基因家族,并從染色體分布、基因及蛋白理化性質(zhì)、蛋白結(jié)構(gòu)、多重序列比對、系統(tǒng)進(jìn)化樹和基因表達(dá)模式等方面對該基因家族的特征進(jìn)行了比較分析。結(jié)果表明,鑒定到37個陸地棉WOX轉(zhuǎn)錄因子家族基因,命名為GhWOX1~GhWOX37。亞基因組定位結(jié)果顯示,37個陸地棉WOX轉(zhuǎn)錄因子家族基因分布在除了A04、A06、A09、D04、D06和D09號亞基因組以外的20個亞基因組,A亞組比D亞組多1個GhWOX轉(zhuǎn)錄因子家族基因。多重序列比對分析棉花WOX轉(zhuǎn)錄因子家族成員均具有高度保守的同源異型結(jié)構(gòu)域;系統(tǒng)進(jìn)化樹分析發(fā)現(xiàn)棉花WOX轉(zhuǎn)錄因子家族可以分為3個亞家族(Ⅰ類、Ⅱ類和Ⅲ類),各亞類分別含有23,7,7個GhWOX轉(zhuǎn)錄因子家族基因成員。對NCBI中陸地棉的EST數(shù)據(jù)庫比對分析,有11個GhWOX轉(zhuǎn)錄因子家族成員沒有可匹配的EST序列,其他26個GhWOX轉(zhuǎn)錄因子家族成員在棉花的根、莖、葉、花、胚珠、纖維和種子等組織和器官中廣泛表達(dá)。為深入研究棉花和其他植物中WOX家族的鑒定和功能研究奠定了基礎(chǔ)。
[Abstract]:In order to mine candidate genes for the study of cotton floral organ or fiber development, the transcription factor gene family of WOX (WUSCHEL-related homeobox,WOX) in upland cotton was identified by bioinformatics based on published genome data of upland cotton. The characteristics of the gene family were compared and analyzed from the aspects of chromosome distribution, physicochemical properties of gene and protein, protein structure, multiple sequence alignment, phylogenetic tree and gene expression pattern. The results showed that 37 WOX transcription factor family genes were identified and named GhWOX1~GhWOX37.. The results of subgenomic mapping showed that 37 WOX transcription factor family genes of Upland cotton were distributed in 20 subgenomes except A04A06A06A09 D04D06 and D09 subgenomes, and one more GhWOX transcription factor family gene was found in A subgroup than that in D subgroup.
