【摘要】：目的：研究端粒保護(hù)蛋白POT1 (protection of telomeres 1) mRNA在非小細(xì)胞肺癌中的表達(dá)情況。分析端粒保護(hù)蛋白POT1在非小細(xì)胞肺癌中的表達(dá)的改變與非小細(xì)胞肺癌的病理分型、分級(jí)以及臨床之間的關(guān)系。方法：收集60例非小細(xì)胞肺癌組織標(biāo)本及癌旁組織標(biāo)本。實(shí)驗(yàn)分為兩組：非小細(xì)胞肺癌組織組和癌旁組織組。提取各組標(biāo)本的總RNA,通過(guò)半定量逆轉(zhuǎn)錄-聚合酶鏈反應(yīng)法(reverse transcription polymerase chain reaction, RT-PCR)對(duì)POT1 mRNA在不同組織中的表達(dá)情況進(jìn)行檢測(cè)。結(jié)果：端粒保護(hù)蛋白POT1 mRNA在非小細(xì)胞肺癌組織組中的光密度比值為0.16114-0.0469,在癌旁組織組中的光密度比值為0.2632±0.1061,端粒保護(hù)蛋白POT1 mRNA在非小細(xì)胞肺癌組織組中的光密度比值明顯低于癌旁組織,組間差異具有統(tǒng)計(jì)學(xué)意義(P0.05)。端粒保護(hù)蛋白POT1 mRNA的表達(dá)在性別、年齡、是否長(zhǎng)期吸煙、分化程度、淋巴結(jié)是否轉(zhuǎn)移、病理TNM分期EGFR基因突變、KRAS基因突變等的對(duì)比中均無(wú)顯著差異(P0.05)。結(jié)論：端粒保護(hù)蛋白POT1 mRNA在非小細(xì)胞肺癌組織組中的相對(duì)表達(dá)量低于其在癌旁組織組中的表達(dá)量.端粒保護(hù)蛋白POT1與非小細(xì)胞肺癌患者性別、年齡、是否長(zhǎng)期吸煙、分化程度、淋巴結(jié)是否轉(zhuǎn)移、病理TNM分期、EGFR基因突變、KRAS基因突等無(wú)相關(guān)性。
[Abstract]:Objective: to study the expression of POT1 protection of telomeres 1 mRNA in non-small cell lung cancer (NSCLC). Methods: 60 specimens of non-small cell lung cancer (NSCLC) and paracancerous tissues (NSCLC) were collected and divided into two groups: NSCLC tissue group and paracancerous tissue group. The expression of POT1 mRNA in different tissues was detected by reverse transcription polymerase chain reaction.Results: the optical density ratio of telomere protection protein POT1 mRNA in non-small cell lung cancer tissues was 0.16114-0.0469. The ratio of optical density in paracancerous tissues was 0.2632 鹵0.1061, and the ratio of telomeric protective protein POT1 mRNA in non-small cell lung cancer tissues was significantly lower than that in paracancerous tissues. The expression of telomere protection protein POT1 mRNA was found in sex, age, long-term smoking, differentiation, lymph node metastasis. There was no significant difference between EGFR gene mutation and KRAS gene mutation in pathological TNM staging. Conclusion: the relative expression of POT1 mRNA in non-small cell lung cancer is lower than that in paracancerous tissue. Telomere Protective protein POT1 and Sex in patients with Non-Small Cell Lung Cancer, There was no correlation among age, long-term smoking, differentiation, lymph node metastasis and pathological TNM staging.
