【摘要】：目的探索建立高效、可靠的腫瘤骨轉(zhuǎn)移動(dòng)物模型的方法。方法將C57BL/6J雄鼠隨機(jī)分為4組,每組15只;BALB/c雌鼠15只,設(shè)一組。將小鼠前列腺癌細(xì)胞RM-1和乳腺癌細(xì)胞4T1-luc消化、洗滌、計(jì)數(shù)后用生理鹽水重懸至1×105/m L,取100μL,經(jīng)股動(dòng)脈將4T1 luc細(xì)胞注射到BALB/c雌鼠,并通過(guò)活體成像檢驗(yàn)此方法轉(zhuǎn)移成瘤的特異性。將100μL相同濃度的RM-1細(xì)胞懸液分別經(jīng)股動(dòng)脈、髂外動(dòng)脈注射C57雄鼠各15只,比較兩種不同注射方式所需時(shí)間、小鼠存活率,注射21天后將下肢骨HE切片,顯微鏡下觀察并比較兩種方法建立骨轉(zhuǎn)移模型的成瘤率。結(jié)果經(jīng)股動(dòng)脈途徑注射腫瘤細(xì)胞懸液所需的止血時(shí)間為2.53±1.75 min,經(jīng)髂外動(dòng)脈注射方法所需的止血時(shí)間為4.70±1.63 min(P0.05);經(jīng)股動(dòng)脈注射方法的手術(shù)時(shí)間為14.67±2.16 min,經(jīng)髂外動(dòng)脈注射方法的手術(shù)時(shí)間為22.47±3.50 min(P0.05);術(shù)后21 d經(jīng)股動(dòng)脈和髂外動(dòng)脈注射組小鼠的生存率分別為93.3%和66.7%(P0.05);采用兩種方法的腫瘤轉(zhuǎn)移率均為100%;分別經(jīng)股動(dòng)脈和髂外動(dòng)脈注射生理鹽水組和腫瘤細(xì)胞懸浮液組在上述3項(xiàng)指標(biāo)無(wú)顯著差異。活體成像結(jié)果顯示,腫瘤特異性地轉(zhuǎn)移到了股骨和脛骨中。結(jié)論經(jīng)股動(dòng)脈注射方法建立腫瘤骨轉(zhuǎn)移模型的手術(shù)及止血時(shí)間短、小鼠的存活率及腫瘤轉(zhuǎn)移的成功率、特異性高,可作為腫瘤骨轉(zhuǎn)移的新型動(dòng)物模型。
[Abstract]:Objective to establish an efficient and reliable animal model of tumor bone metastases. Methods C57BL / 6J male mice were randomly divided into 4 groups, 15 female mice in each group were divided into one group. The mouse prostate cancer cell line RM-1 and breast cancer cell line 4T1-luc were digested and washed. After counting, 100 渭 L of normal saline was resuspended to 1 脳 105 / mL, and 4T1 luc cells were injected into the female BALB/c rat via femoral artery, and the specificity of the tumor was examined by in vivo imaging. The 100 渭 L RM-1 cell suspension of the same concentration was transferred to the femoral artery, respectively. 15 male C57 rats were injected into the external iliac artery. The time required for two different injection methods and the survival rate of the mice were compared. The lower extremity bone was sliced with HE on 21 days after injection. Results the hemostatic time for injection of tumor cell suspension through femoral artery was 2.53 鹵1.75 min, and the hemostatic time for external iliac artery injection was 4.70 鹵1.63 min (P0.05). The operative time of transfemoral and external iliac artery injection was 14.67 鹵2.16 min and 22.47 鹵3.50 min respectively, and the survival rate was 93.3% and 66.7% P0.05in femoral artery and external iliac artery injection group 21 days after operation, respectively. There was no significant difference in the above three indexes between the saline injection group and the tumor cell suspension group, and the results of in vivo imaging showed that there was no significant difference between the two groups. Conclusion the operative and hemostatic time of tumor bone metastasis model established by femoral artery injection is short, and the survival rate of mice and the success rate of tumor metastasis are high. It can be used as a new animal model of tumor bone metastasis.
