回轉(zhuǎn)模擬失重通過激活miR-103/Cav1.2信號通路抑制MC3T3-E1前成骨細(xì)胞增殖的研究

研究背景骨密度的保持和骨骼結(jié)構(gòu)的發(fā)育依賴于機(jī)械力學(xué)刺激。研究表明,機(jī)械力學(xué)刺激在骨重塑過程中促進(jìn)骨形成,然而在長期制動或是失重環(huán)境下的去負(fù)荷狀態(tài)導(dǎo)致骨質(zhì)丟失。失重環(huán)境暴露可引起航天員各系統(tǒng)功能失調(diào),其中最為嚴(yán)重的是非自限性骨質(zhì)丟失,這將會增加航天員發(fā)生骨折的危險。長時間的失重暴露使骨質(zhì)生成減少,骨質(zhì)吸收增加。失重誘導(dǎo)的骨質(zhì)丟失能夠以骨密度每個月接近2%的速度下降,此種去負(fù)荷環(huán)境作用下一個月的骨質(zhì)丟失量和絕經(jīng)期婦女一年的骨質(zhì)丟失量相當(dāng)。研究已證實,成骨細(xì)胞增殖能力的減弱在失重性骨質(zhì)丟失的過程中扮演重要角色,但具體機(jī)制尚不清楚。研究表明,鈣離子是成骨細(xì)胞的重要調(diào)節(jié)因子,而鈣離子通道,特別是L型電壓依賴型鈣離子通道(L-type calcium channels,LTCCs)在機(jī)械刺激誘導(dǎo)的成骨細(xì)胞增殖中發(fā)揮重要作用。一些證據(jù)表明,當(dāng)LTCCs被激活時,骨密度增加伴隨骨溶解減少。應(yīng)用特異性LTCCs阻斷劑,維拉帕米或nifedipine作用于成骨細(xì)胞可阻斷機(jī)械刺激誘導(dǎo)的成骨細(xì)胞增殖。另有研究表明,機(jī)械刺激能夠促進(jìn)成骨細(xì)胞細(xì)胞外基質(zhì)分泌增加,而應(yīng)用特異性LTCCs阻斷劑后此效應(yīng)消失。所以LTC...
