【摘要】：目的建立一種快速檢測(cè)琥珀膽堿的方法。方法血液樣品經(jīng)過乙腈沉淀蛋白后,以0.22μm濾膜過濾,利用高效液相色譜-飛行時(shí)間質(zhì)譜聯(lián)用儀檢測(cè)毒狗針及狗血中琥珀膽堿成分。選用Kinetex 2.6μm HILIC(100mm×2.1 mm)并前接保護(hù)柱,以5 mmol醋酸銨90%乙腈水溶液和200 mmol醋酸銨0.4%甲酸水溶液作為流動(dòng)相,進(jìn)行等度洗脫分離。結(jié)果前處理簡(jiǎn)單快速,檢測(cè)時(shí)間僅需5 min。采用的定性定量離子其精確分子量為145.109 3Da。測(cè)得血液樣品中琥珀膽堿的檢測(cè)限為0.9 ng/m L(S/N=10),方法的線性范圍為0.01~10μg/m L。結(jié)論該方法準(zhǔn)確可靠、簡(jiǎn)便快速,具有良好重現(xiàn)性,是琥珀膽堿的一種有效檢測(cè)方法。
[Abstract]:Objective to establish a rapid method for the determination of succinylcholine. Methods the blood samples were precipitated by acetonitrile and filtered by 0.22 渭 m filter membrane. The succinylcholine in the venomous dog needle and dog blood was determined by high performance liquid chromatography-time-of-flight mass spectrometry (HPLC / MS). Kinetex 2.6 渭 m HILIC (100mm 脳 2. 1 mm) was used as the mobile phase with 5 mmol ammonium acetate 90% acetonitrile solution and 200 mmol ammonium acetate 0.4% formic acid solution as the mobile phase. Results the pretreatment was simple and rapid, and the detection time was only 5 min. The accurate molecular weight of the qualitative and quantitative ions is 145.109 3 Da. The detection limit of succinylcholine in blood samples was 0.9 ng/m / L (S / N ~ (10). The linear range of the method was 0.01 ~ 10 渭 g / m ~ (-1). Conclusion the method is accurate, simple, rapid and reproducible. It is an effective method for the determination of succinylcholine.
