肉類摻偽的毛細(xì)管電泳檢測(cè)法

【摘要】:目的建立肉類的PCR-CE-LIF檢測(cè)方法,并用以檢測(cè)市售肉類的摻偽。方法采用DNA Ladder Marker(100 bp)優(yōu)化CE的分離檢測(cè)條件,包括分離電壓、篩分介質(zhì)濃度和電泳緩沖液濃度。采用試劑盒法提取肉的基因組DNA,并用特異性引物對(duì)其特異保守序列進(jìn)行PCR擴(kuò)增,經(jīng)HindⅢ限制性核酸內(nèi)切酶酶切,產(chǎn)物用優(yōu)化的CE-LIF條件檢測(cè),測(cè)定結(jié)果與瓊脂糖凝膠電泳法進(jìn)行比對(duì)。結(jié)果在優(yōu)化的CE條件下,同一天連續(xù)6次測(cè)定DNA Ladder Marker,遷移時(shí)間的日內(nèi)相對(duì)標(biāo)準(zhǔn)差(RSD)為0.21%~0.71%;連續(xù)6天測(cè)定,遷移時(shí)間的日間RSD為1.02%~2.09%。毛細(xì)管電泳法能特異、靈敏地鑒定不同肉類,而瓊脂糖凝膠電泳則無(wú)法檢出目的條帶。結(jié)論 CE-LIF方法具有高靈敏度、高分辨率、高特異性、綠色環(huán)保等優(yōu)勢(shì),能夠用于肉類摻偽測(cè)定。
[Abstract]:Objective to establish a PCR-CE-LIF method for detection of meat adulteration. Methods the separation and detection conditions of CE were optimized by DNA ladder Marker (100 BP), including the separation voltage, the concentration of screening medium and the concentration of electrophoretic buffer.
