蘋果葉總黃酮的提取及其含量測定

【摘要】:以"紅元帥"蘋果的干燥葉子為試材,通過單因素試驗確定蘋果葉總黃酮的最佳提取工藝條件,采用可見分光光度法對蘋果葉中總黃酮含量進(jìn)行測定,以期為蘋果葉中總黃酮提取工藝及含量測定提供方法和依據(jù)。結(jié)果表明:蘋果葉總黃酮的最佳提取工藝條件為乙醇作溶劑,乙醇濃度60%,料液比1∶20g/mL,提取溫度為70℃,提取時間為30min;在選定試驗條件下對蘋果葉進(jìn)行含量測定,對蘆丁對照品濃度與吸光度進(jìn)行線性回歸,其回歸方程為Y=24.821 43X-0.031,R2=0.999 45(n=6),蘆丁對照品質(zhì)量在4~24μg范圍內(nèi)呈良好的線性關(guān)系,在此條件下測定蘋果葉中總黃酮平均含量為30.28mg/g。試驗優(yōu)選出了蘋果葉中總黃酮的最佳提取工藝條件及含量測定方法,該方法簡單、快速,準(zhǔn)確,精密度,穩(wěn)定性,重現(xiàn)性良好。
[Abstract]:Using dried leaves of "Red Marshal" apple as test material, the optimum extraction conditions of total flavonoids from apple leaves were determined by single factor test, and the content of total flavonoids in apple leaves was determined by visible spectrophotometry.In order to provide a method and basis for the extraction and determination of total flavonoids in apple leaves.The results showed that the optimum conditions of extracting total flavonoids from apple leaves were ethanol as solvent, ethanol concentration as 60%, ratio of material to liquid 1: 20 g / mL, extraction temperature 70 鈩,
