【摘要】：目的建立流動(dòng)注射化學(xué)發(fā)光法測(cè)定血漿中奈替米星的方法。方法在堿性介質(zhì)中,奈替米星對(duì)鐵氰化鉀與魯米諾化學(xué)發(fā)光體系具有明顯的增敏作用,據(jù)此建立了一種測(cè)定奈替米星的流動(dòng)注射化學(xué)發(fā)光新方法,對(duì)分析條件進(jìn)行優(yōu)化。通過(guò)方法學(xué)評(píng)價(jià)及對(duì)血漿奈替米星的測(cè)定,觀察建立方法的性能。結(jié)果優(yōu)化后的最佳檢測(cè)條件是:魯米諾濃度為10μmol/L,鐵氰化鉀的濃度為30μmol/L,Na OH的濃度為0.10mol/L,采樣時(shí)間為5s,泵速為30r/min,負(fù)高壓為630V。在確定的最佳實(shí)驗(yàn)條件下,化學(xué)發(fā)光增加強(qiáng)度與奈替米星的濃度在1.0×10~(-6)~5.0×10~(-5)g/m L范圍內(nèi)呈良好的線性關(guān)系,檢測(cè)限為9.2×10~(-7)g/m L。對(duì)含3.6×10~(-6)g/m L奈替米星的溶液平行測(cè)定11次,測(cè)定結(jié)果的相對(duì)標(biāo)準(zhǔn)偏差為2.3%。結(jié)論本方法分析速度快、靈敏度高、分析成本低,可用于血漿中奈替米星的測(cè)定。
[Abstract]:Objective to establish a flow injection chemiluminescence method for the determination of netilmicin in plasma. Methods in alkaline medium, netilmicin had obvious sensitizing effect on potassium ferricyanide and luminol chemiluminescence system. A new flow injection chemiluminescence method for the determination of netilmicin was established and the analytical conditions were optimized. The performance of the established method was observed by methodological evaluation and determination of netilmicin in plasma. Results the optimum detection conditions were as follows: the concentration of luminol was 10 渭 mol / L, the concentration of potassium ferricyanide was 30 渭 mol / L, the concentration of Na OH was 0.10 mol 路L, the sampling time was 5 s, the pump speed was 30 r / min, and the negative pressure was 630 V. Under the optimum experimental conditions, there is a good linear relationship between the intensity of chemiluminescence and the concentration of netilmicin in the range of 1.0 脳 10 ~ (- 6) ~ 5.0 脳 10 ~ (- 5) g 路mL, and the detection limit is 9.2 脳 10 ~ (- 7) g 路mL ~ (- 1). The relative standard deviation of the solution containing 3.6 脳 10 ~ (- 6) g 脳 10 ~ (- 6) g / L netilmicin was 2.3%. Conclusion this method has the advantages of fast analysis speed, high sensitivity and low analysis cost. It can be used for the determination of netilmicin in plasma.
