星點設計-響應面法優(yōu)化穿膜肽修飾紫杉醇和他莫昔芬脂質(zhì)體的處方

【摘要】:目的:采用響應面法優(yōu)選出制備穿膜肽(CPP)修飾紫杉醇和他莫昔芬脂質(zhì)體的最佳處方。方法:采用薄膜分散-硫酸銨梯度法制備CPP修飾紫杉醇和他莫昔芬脂質(zhì)體,以磷脂/膽固醇(摩爾比),磷脂/聚乙二醇-二硬脂酰磷脂酰乙醇胺(摩爾比),磷脂/紫杉醇(摩爾比)為自變量,以修飾紫杉醇和他莫昔芬包封率的綜合評分為因變量進行二次多項擬合。結果:脂質(zhì)體的最優(yōu)處方為磷脂/膽固醇=50:1,磷脂/PEG 2000-DSPE=5.5:1,磷脂/紫杉醇=50:1,脂質(zhì)體中紫杉醇和他莫昔芬含量的綜合評分為95.48%。結論:星點設計-響應面法優(yōu)選出了CPP修飾紫杉醇和他莫昔芬脂質(zhì)體的最佳處方。
[Abstract]:Aim: to optimize the preparation of paclitaxel and tamoxifen liposomes modified by transmembrane peptide (CPP) by response surface method. Methods: CPP modified paclitaxel and tamoxifen liposomes were prepared by thin film dispersion ammonium sulfate gradient method with phospholipid / cholesterol (molar ratio), phospholipid / polyethylene glycol distearyl phosphatidylethanolamine (molar ratio). Phospholipid / paclitaxel (molar ratio) was used as independent variable, and the comprehensive score of entrapment efficiency of modified paclitaxel and tamoxifen was used as dependent variable for quadratic multivariate fitting.
