液相色譜-四極桿飛行時(shí)間質(zhì)譜檢測染毒動(dòng)物血漿中沙林和梭曼酪氨酸加合物

【摘要】:為了追溯性檢測神經(jīng)性毒劑沙林(GB)和梭曼(GD)暴露染毒,以染毒白蛋白上的特異性加合位點(diǎn)-411位酪氨酸加合物為重要生物標(biāo)志物,建立了高效液相色譜-四極桿飛行時(shí)間質(zhì)譜(LC/Q-TOF MS)分析動(dòng)物血液中神經(jīng)性毒劑GB和GD酪氨酸加合物的檢測方法。在采用不同樣品處理和儀器條件的基礎(chǔ)上,確定了最佳實(shí)驗(yàn)條件。血液經(jīng)離心后,取50μL分離的血漿加入到50μL 10 g/L鏈霉蛋白酶的NH4HCO3(50 mmol/L)水溶液中,于37℃孵育2.5 h,用0.5 m L 10 ku超濾管以16 000 r/min離心15min,LC/Q-TOF MS電噴霧正離子模式分析濾液。應(yīng)用本方法進(jìn)行GB半致死劑量(LD50)動(dòng)物施毒實(shí)驗(yàn),在施毒的動(dòng)物血樣中檢測到膦酰化酪氨酸加合物,證明了該方法快速、簡單、可靠。
