【摘要】：目的:探討精神類藥物舍曲林對(duì)人急性髓系白血病細(xì)胞株Kasumi-1的生物學(xué)效應(yīng)。方法:將不同濃度舍曲林作用于Kasumi-1細(xì)胞不同時(shí)間,通過細(xì)胞計(jì)數(shù)、細(xì)胞形態(tài)學(xué)檢查、流式細(xì)胞術(shù)和Western blot分別檢測(cè)舍曲林處理前后Kasumi-1細(xì)胞增殖和凋亡情況、細(xì)胞周期分布以及相關(guān)蛋白的改變。結(jié)果:舍曲林能夠抑制Kasumi-1細(xì)胞增殖、誘導(dǎo)細(xì)胞凋亡;15和20μmol/L舍曲林處理Kasumi-1細(xì)胞24 h后,生長(zhǎng)抑制率分別達(dá)到(19.00±7.37)%和(47.90±11.19)%;流式細(xì)胞術(shù)檢測(cè)結(jié)果顯示,與空白對(duì)照組相比,15和20μmol/L舍曲林處理Kasumi-1細(xì)胞24 h后,Annexin V陽(yáng)性細(xì)胞百分比從(9.71±2.12)%分別上升至(20.54±2.52)%和(45.37±7.88)%(P0.01),且細(xì)胞被明顯阻滯在G0/G1期和G2/M期;此外,舍曲林還可以下調(diào)細(xì)胞內(nèi)翻譯控制腫瘤蛋白(translationally controlled tumor protein,TCTP)。結(jié)論:舍曲林能夠顯著誘導(dǎo)Kasumi-1細(xì)胞發(fā)生凋亡,其作用機(jī)制可能與細(xì)胞內(nèi)TCTP蛋白的下調(diào)相關(guān)。
[Abstract]:Objective: to investigate the biological effects of sertraline on human acute myeloid leukemia cell line Kasumi-1. Methods: Kasumi-1 cells were treated with Sertraline at different concentrations for different time. Cell count, cell morphology, flow cytometry and Western blot were used to detect the proliferation and apoptosis of Kasumi-1 cells before and after sertraline treatment. Cell cycle distribution and changes in related proteins. Results: Sertraline inhibited the proliferation of Kasumi-1 cells and induced apoptosis, and the growth inhibition rates of Kasumi-1 cells treated with sertraline for 24 h were (19.00 鹵7.37)% and (47.90 鹵11.19)%, respectively. The results of flow cytometry showed that Kasumi-1 cells were treated with 15 and 20 渭 mol/L sertraline for 24 h, compared with the control group. The percentage of Annexin V positive cells increased from (9.71 鹵2.12)% to (20.54 鹵2.52)% and (45.37 鹵7.88)%, respectively. In addition, sertraline can also down-regulate intracellular translation control tumor protein (translationally controlled tumor protein,TCTP). Conclusion: Sertraline can induce apoptosis in Kasumi-1 cells, and its mechanism may be related to the down-regulation of TCTP protein.
