【摘要】：成纖維細胞生長因子受體4(FGFR4)通過啟動STAT3、MAPK和PI3K/AKT等信號級聯(lián),調(diào)節(jié)細胞的增殖、分化和轉(zhuǎn)移。其基因突變、過表達或其配體分子過表達等異常信號轉(zhuǎn)導在某些腫瘤的發(fā)生發(fā)展中發(fā)揮著重要的作用,表明FGFR4是此類腫瘤潛在的治療靶標。靶向FGFR4的抗腫瘤藥物或方法主要包括小分子抑制劑、單克隆抗體、配體捕獲蛋白、短鏈RNA寡核苷酸適配體(shRNA)。本文回顧FGFR4異常的腫瘤及靶向FGFR4抗腫瘤藥物的研究進展。
[Abstract]:Fibroblast growth factor receptor 4 (FGFR4) regulates cell proliferation, differentiation and metastasis by activating signal cascades such as STAT3,MAPK and PI3K/AKT. Abnormal signal transduction, such as gene mutation, overexpression or overexpression of its ligand, plays an important role in the occurrence and development of some tumors, indicating that FGFR4 is a potential therapeutic target for these tumors. Antitumor drugs or methods targeting FGFR4 include small molecular inhibitors, monoclonal antibodies, ligand capping proteins, and short chain RNA oligonucleotide aptamers, (shRNA). This review reviews the research progress of FGFR4 abnormal tumors and FGFR4-targeted anti-tumor drugs.
