大鼠基底外側(cè)杏仁核組蛋白乙;瘜岱瘸砂a記憶的調(diào)控作用

【摘要】:為了探索組蛋白乙;瘜岱瘸砂a記憶相關(guān)分子表達(dá)調(diào)控機(jī)制,文章選取健康成年雄性SD大鼠34只,隨機(jī)分為正常對照組(n=6)及基底外側(cè)杏仁核(Basolateral amygdala,BLA)顱內(nèi)定位手術(shù)組(n=28)。在條件性位置偏愛(Conditioned place preference,CPP)訓(xùn)練階段,大鼠BLA內(nèi)給予組蛋白去乙;敢种苿┣乓志谹(Trichostafin A,TSA)并且腹腔注射嗎啡溶液(10.0 mg/kg),對照組給予相同體積的10%二甲基亞砜(Dimethyl sulfoxide,DMSO)或鹽水。應(yīng)用蛋白質(zhì)印記方法,檢測嗎啡誘導(dǎo)大鼠CPP建立后BLA內(nèi)組蛋白H3K14乙;湍X源性神經(jīng)營養(yǎng)因子(Brain-derived neurotrophic factor,BDNF)蛋白表達(dá)水平。結(jié)果顯示,腹腔注射10 mg/kg嗎啡能成功建立CPP。嗎啡、TSA聯(lián)合給藥組大鼠比單純嗎啡給藥組大鼠表現(xiàn)出更強(qiáng)烈的CPP(P0.0001)。嗎啡和TSA都能使BLA內(nèi)的組蛋白H3乙;胶虰DNF的表達(dá)顯著增高(P0.0001),同時二者之間具有協(xié)同作用。結(jié)果表明,大鼠BLA內(nèi)組蛋白乙;脚c嗎啡成癮記憶形成有關(guān),抑制BLA內(nèi)組蛋白去乙酰化酶(Histone deacetylases,HDACs)的活性可強(qiáng)化嗎啡誘導(dǎo)的線索記憶的形成;大鼠BLA內(nèi)BDNF參與了嗎啡誘導(dǎo)的線索記憶的形成并可能受到組蛋白乙;恼{(diào)控。
[Abstract]:In order to explore the regulation mechanism of histone acetylation on the expression of memory related molecules in morphine addiction, 34 healthy male SD rats were randomly divided into normal control group (nong6) and basal lateral amygdaloid nucleus (Basolateral amygdala,). BLA) Intracranial localization operation group (NM 28). During conditioned place preference (Conditioned place preference,CPP) training stage, rats were given histone deacetylase inhibitor (A (Trichostafin) and intraperitoneal injection of morphine solution (10.0 mg/kg) in BLA. The control group was given the same volume of 10% dimethyl sulfoxide (Dimethyl sulfoxide,DMSO) or saline. The acetylation of histone H3K14 and the expression of brain-derived neurotrophic factor (Brain-derived neurotrophic factor,BDNF) in BLA of morphine induced rat CPP were detected by protein imprinting method. The results showed that CPP. could be successfully established by intraperitoneal injection of morphine for 10 mg/kg. The CPP of morphine, TSA combined administration group was stronger than that of morphine alone group (P0.0001). Both morphine and TSA could significantly increase the level of histone H 3 acetylation and the expression of BDNF in BLA (P0.0001), and there was a synergistic effect between them. The results showed that the level of histone acetylation in rat BLA was related to the formation of morphine addictive memory. Inhibiting the activity of histone deacetylase (Histone deacetylases,HDACs) in BLA could enhance the formation of morphine induced clue memory. BDNF in rat BLA is involved in the formation of clue memory induced by morphine and may be regulated by histone acetylation.
