【摘要】：目的建立1種同時(shí)測(cè)定骨瓜提取物注射劑中15種氨基酸含量的RP-HPLC法。方法以異硫氰酸苯酯(PITC)為柱前衍生試劑對(duì)樣品中15種氨基酸進(jìn)行衍生化,采用Ocean C18(250 mm×4.6 mm,5μm)色譜柱,流動(dòng)相A為水-乙腈(體積比1∶4),流動(dòng)相B為濃度0.1 mol·L-1乙酸鈉-乙腈(體積比97∶3),梯度洗脫,流速為1.0 m L·min-1,柱溫為25℃,檢測(cè)波長(zhǎng)為254 nm,進(jìn)樣量為10μL。用內(nèi)標(biāo)法計(jì)算15種氨基酸的含量。結(jié)果 15種氨基酸質(zhì)量濃度在1.24~160.72 mg·L-1內(nèi)與峰面積呈良好的線性關(guān)系(r=0.997 7~1.000 0),15種氨基酸的平均加樣回收率在97.7~102.4%內(nèi),加樣回收率的RSD值在1.7%~3.3%內(nèi)。結(jié)論該方法準(zhǔn)確、可靠、靈敏,可以用于同時(shí)測(cè)定骨瓜提取物注射劑中15種氨基酸的含量。
[Abstract]:Objective to establish a RP-HPLC method for simultaneous determination of 15 amino acids in extract injection of Gugu melon.
