殼寡糖對HeLa細胞和A549細胞自噬性死亡的影響

【摘要】:為了探討殼寡糖(COS)對He La細胞和A549細胞自噬性死亡的影響,利用不同濃度COS處理細胞后MTT法檢測細胞的增殖抑制情況,采用MDC染色法觀察自噬泡的形成,免疫熒光法檢測LC3在細胞內(nèi)的表達,Western-blot分析COS處理He La和A549細胞不同時間后細胞內(nèi)LC3-Ⅱ/LC3-Ⅰ變化。結果表明,5.0 mg·m L-1COS對兩種細胞的增殖抑制率最大,分別為52.15%和57.63%(P0.01)。與陰性對照相比,經(jīng)COS處理后的細胞MDC染色細胞內(nèi)熒光強度增加,且陽性信號聚集于核周區(qū)域;免疫熒光檢測發(fā)現(xiàn)LC3陽性顆粒數(shù)明顯增加,表明細胞內(nèi)自噬體的數(shù)量增多。Western-blot檢測分析隨著COS處理細胞時間延長,LC3-Ⅱ/LC3-Ⅰ比值逐漸增大,顯示細胞內(nèi)自噬活性增強。由此推測COS能夠通過誘導He La和A549細胞的自噬性死亡來抑制細胞增殖,為COS的抗腫瘤作用機制的研究提供理論依據(jù)。
