【摘要】：目的觀察D-檸檬烯對(duì)酒精性腦損傷大鼠腦組織NF-κB、IκB及TNF-α表達(dá)的影響。方法 60只Wistar大鼠隨機(jī)分為6組,每組各10只。藥物1、2、3組與模型組制作酒精性腦損傷模型,每日予50%乙醇灌胃,前2周為8 m L/(kg·d),后4周為12 m L/(kg·d),其中藥物1、2、3組造模同時(shí)分別予100、200、400 mg/(kg·d)的D-檸檬烯灌胃;單純藥物組與空白對(duì)照組不制作動(dòng)物模型,其中空白對(duì)照組每日予蒸餾水灌胃,單純藥物組在此基礎(chǔ)上加400 mg/(kg·d)D-檸檬烯灌胃。實(shí)驗(yàn)第6周末處死動(dòng)物,留取腦組織,HE染色,鏡下觀察其形態(tài)結(jié)構(gòu)變化;免疫組化法測(cè)定腦組織中的NF-κB、IκB蛋白;ELISA雙抗夾心法檢測(cè)腦勻漿中的TNF-α。結(jié)果模型組神經(jīng)元數(shù)量減少,分布不規(guī)則,細(xì)胞核明顯固縮變形,藥物1、2、3組腦組織形態(tài)結(jié)構(gòu)變化逐漸減輕。模型組NF-κB陽(yáng)性細(xì)胞數(shù)、IκB陽(yáng)性細(xì)胞數(shù)與空白對(duì)照組相比,P均0.05。藥物1、2、3組NF-κB陽(yáng)性細(xì)胞數(shù)減少、IκB陽(yáng)性細(xì)胞數(shù)增多,與模型組相比,P均0.05;藥物3組與藥物1、2組相比,P均0.05。模型組大鼠腦勻漿中TNF-α表達(dá)高于空白對(duì)照組(P均0.05);藥物1、2、3組大鼠腦勻漿中TNF-α表達(dá)低于模型組(P均0.05),且藥物3組低于藥物1、2組(P均0.05)。結(jié)論 D-檸檬烯可降低酒精性腦損傷大鼠腦組織中NF-κB、TNF-α的表達(dá),上調(diào)IκB的表達(dá),進(jìn)而減輕酒精性腦損傷。
[Abstract]:Objective to observe the effect of D-limonene on the expression of NF- 魏 B I 魏 B and TNF- 偽 in brain tissue of rats with alcoholic brain injury. Methods 60 Wistar rats were randomly divided into 6 groups with 10 rats in each group. The model of alcoholic brain injury was established in group 1 and group 2. The rats were given 50% ethanol daily, 12 mL / (kg d), for the first 2 weeks and 8 mL / (kg d), for 4 weeks. The rats in group 1 and 2 were given 100200400 mg/ (kg d) of D-limonene by stomach at the same time. There were no animal models in the drug group and the blank control group, in which the blank control group was fed with distilled water daily, and the simple drug group was fed with 400 mg/ (kg d) D-limonene. The animals were killed at the end of the 6th week of the experiment, the brain tissue was stained with HE and the changes of morphology and structure were observed under microscope, and the content of TNF- 偽 in brain homogenate was detected by Elisa double antibody sandwich method. Results in the model group, the number of neurons decreased, the distribution of neurons was irregular, the nucleus was obviously constricted and deformed. The number of NF- 魏 B positive cells and the number of I 魏 B positive cells in the model group were 0.05 compared with those in the blank control group. The number of NF- 魏 B positive cells in drug 1 + 2 + group decreased, the number of I 魏 B positive cells increased, compared with model group (P 0.05), drug 3 group (P < 0.05) compared with drug 1 "2" group (P < 0. 05). The expression of TNF- 偽 in the brain homogenate of the model group was higher than that in the blank control group (all P 0.05), and the expression of TNF- 偽 in the brain homogenate of the model group was lower than that in the model group (P 0.05), and the expression of TNF- 偽 in the brain homogenate of the drug group was lower than that in the control group (all P 0.05). Conclusion D- limonene can decrease the expression of NF- 魏 B TNF- 偽 and up-regulate the expression of I 魏 B in brain tissue of rats with alcoholic brain injury, and then attenuate the brain injury induced by alcohol.
