溫控型胰島素液體肛門栓的研究

【摘要】:目的:溫控型胰島素液體肛門栓的研究,其內(nèi)容包括含量測定方法、制備工藝、體外釋放、穩(wěn)定性、藥動學(xué)和藥效學(xué)。方法:建立HPLC-UV測定溫控型胰島素液體肛門栓中胰島素含量的方法。色譜柱為Welch ultimte C18,流動相為0.2mol·L-1硫酸鹽緩沖液-乙腈(74:26,V:V);流速1.0mL.min-1檢測波長214nm;柱溫40℃;進樣量20μL。運用正交試驗以形成凝膠的時間作為考察指標(biāo)來確定溫控型胰島素液體肛門栓的基質(zhì)制備工藝。利用無膜透析試驗考察溫控型胰島素液體肛門栓的凝膠溶蝕情況,利用透析袋試驗考察溫控型胰島素液體肛門栓的分子量對進入人體的影響,以牛、羊腸黏膜釋放試驗?zāi)M考察人腸黏膜對藥物的吸收情況。制備溫控型胰島素液體肛門栓,進行高溫試驗、高濕度試驗、強光照射試驗,考察溫控型胰島素液體肛門栓的穩(wěn)定性。采用測定血清胰島素的方法研究糖尿病兔體內(nèi)的藥動學(xué)過程,計算主要藥動學(xué)參數(shù)。通過血糖值的測定考察溫控型胰島素液體肛門栓對正常家兔和糖尿病兔的降血糖作用。結(jié)果:經(jīng)方法學(xué)考察,含量測定方法簡便、準(zhǔn)確,溫控型胰島素液體肛門栓在1.0~40.0IU范圍內(nèi)線性關(guān)系良好(r=0.9991),平均回收率99.35%,RSD0.79%。通過正交試驗,確定最佳制備工藝為P407和P188比例15:25,殼聚糖0.4%,pH值5±0.2。無膜釋放:Q=0.4954t-2.6118(r=0.9977);牛腸黏膜釋放:(1-Q)1/3=0.0016t+2.9274(r=-0.9755)羊腸黏膜釋放:lnQ=0.8783lnt-4.931(r=0.9705);透析袋釋放1nQ=0.23lnt-2.1936(r=0.9839),體外釋放研究試驗結(jié)果表明胰島素是通過擴散和溶蝕作用進入體內(nèi)的。主要藥動學(xué)參數(shù):Tmax低、中、高劑量組:(90.0±21.2).(90.0±18.9).(78.0±24.0)min;Cmax低、中、高劑量組:(109.7±36.6)、(115.2±28.1)、(151.3±40.3)μIU·mL-1;相對生物利用度低、中、高劑量組:32.86%、20.20%、15.31%。不同劑量溫控型胰島素液體肛門栓可降低糖尿病家兔的血糖值、升高血清胰島素濃度。結(jié)論:溫控型胰島素液體肛門栓給藥后各劑量組糖尿病兔血糖下降、血清胰島素含量上升與生理鹽水組比較有顯著性差異P0.05。溫控型胰島素液體肛門栓可由直腸黏膜吸收發(fā)揮作用。
