豚鼠胰腺內(nèi)telocytes形態(tài)學(xué)特點(diǎn)及甲磺酸伊馬替尼對(duì)telocytes的干預(yù)作用

【摘要】:目的對(duì)豚鼠胰腺組織特絡(luò)細(xì)胞(telocytes,TCs)的形態(tài)特征進(jìn)行觀察與分析,并探討甲磺酸伊馬替尼對(duì)TCs的干預(yù)作用。方法以健康9周齡豚鼠胰腺為研究對(duì)象,利用免疫組化染色對(duì)TCs免疫標(biāo)志(c-kit/CD117、vimentin)進(jìn)行觀察分析。透射電鏡觀察TCs的超微結(jié)構(gòu)。建立甲磺酸伊馬替尼干預(yù)模型:豚鼠54只,雌雄不限,分正常對(duì)照組、用藥組(7、14、21、28 d)、停藥組(W7、W14、W21、W28 d);免疫組化染色觀察用/停藥后TCs變化;Western blot觀察用/停藥后胰腺組織內(nèi)CD117、vimentin蛋白表達(dá)。結(jié)果免疫組化染色顯示:胰腺內(nèi)TCs位于結(jié)締組織內(nèi),細(xì)胞呈現(xiàn)三角形及長(zhǎng)梭形等多種形態(tài),有一至多條長(zhǎng)而細(xì)的細(xì)胞突起,突起包繞腺泡,并相連形成網(wǎng)絡(luò)結(jié)構(gòu);電鏡超微結(jié)構(gòu)顯示:TCs有念珠狀的細(xì)長(zhǎng)突起,核質(zhì)比較高,胞質(zhì)內(nèi)含有少量的線粒體、滑面內(nèi)質(zhì)網(wǎng)等;甲磺酸伊馬替尼干預(yù)后,TCs突起變短,網(wǎng)絡(luò)結(jié)構(gòu)減少,停藥后逐漸恢復(fù);用藥后胰腺組織內(nèi)c-kit/CD117和vimentin蛋白表達(dá)降低,停藥后逐漸恢復(fù),差異有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論豚鼠胰腺內(nèi)存在梭形有念珠狀突起的TCs,TCs包繞腺泡形成網(wǎng)絡(luò)結(jié)構(gòu);甲磺酸伊馬替尼可影響胰腺內(nèi)TCs的形態(tài)及蛋白表達(dá)。
[Abstract]:Objective to observe and analyze the morphologic characteristics of telocytes-TCs in guinea pig pancreas and to investigate the effect of imatinib mesylate on TCs. Methods Nine-week-old guinea pig pancreas was used to observe and analyze TCs immunological markers (c-kit / CD _ (117) vimentin) by immunohistochemical staining. The ultrastructure of TCs was observed by transmission electron microscope. The model of imatinib mesylate intervention was established: 54 guinea pigs, male and female, were divided into two groups: normal control group and control group. The changes of TCs were observed by immunohistochemical staining of TCs in the treated group (W _ (14) W _ (14) W _ (21) W _ (21) and W _ (28) d); and the expression of CD117vimentin protein in pancreatic tissue was observed by Western blot. Results Immunohistochemical staining showed that TCs in the pancreas was located in connective tissue. The cells were triangular and fusiform. There were one or more long and thin processes around the acinar and connected to form the network structure. The ultrastructure of electron microscope showed that TCs had the slender bulge of rosary, the nucleus and cytoplasm were relatively high, the cytoplasm contained a small amount of mitochondria and the smooth endoplasmic reticulum, etc. After the intervention of imatinib mesylate, the process of TCs became shorter, the network structure decreased, and gradually recovered after drug withdrawal. After treatment, the expression of c-kit/CD117 and vimentin protein in pancreatic tissue decreased, and gradually recovered after withdrawal, the difference was statistically significant (P0.05). Conclusion in the pancreas of guinea pig, the spindle shape and the rosary protuberance form the network structure around the acinar, and imatinib mesylate affects the morphology and protein expression of TCs in the pancreas.
