白藜蘆醇衍生物的合成及其初步細胞活性研究

【摘要】:目的:合成白藜蘆醇的衍生物--乙酰白藜蘆醇和甲基化白藜蘆醇,并對其進行結(jié)構(gòu)解析;建立它們的液相色譜測定含量方法并測定脂水分配系數(shù);觀察它們對3T3-L1脂肪細胞的影響。 方法:以白藜蘆醇為原料,通過化學(xué)合成乙酰白藜蘆醇和甲基化白藜蘆醇,以核磁共振H譜、C譜、紫外光譜與質(zhì)譜對得到的產(chǎn)物進行結(jié)構(gòu)解析;以高效液相色譜法測定含量和脂水分配系數(shù);細胞實驗部分,實驗分為空白對照組、白藜蘆醇對照組、甲基化白藜蘆醇組、乙酰白藜蘆醇組,用MTT法測定不同濃度的甲基化白藜蘆醇和乙酰白藜蘆醇對3T3-L1脂肪細胞增殖的抑制作用;甲基化白藜蘆醇、乙酰白藜蘆醇分別處理24h和48h后,采用AnnexinV-PI雙染法檢測定其誘導(dǎo)3T3-L1脂肪細胞細胞凋亡作用,采用RT-PCR法檢測定其對3T3-L1脂肪細胞中PPARγ的mRNA表達的影響。 結(jié)果:通過結(jié)構(gòu)確認,合成得到了乙酰白藜蘆醇和甲基化白藜蘆醇,,純度均大于99%;其脂水分配系數(shù)(25℃)分別為:LogP=1.49,4.20;細胞實驗部分,與空白對照組比較,甲基化白藜蘆醇組、乙酰白藜蘆醇組均對3T3-L1脂肪細胞的細胞活力有明顯的抑制作用,且呈濃度時間依賴關(guān)系,其IC50分別為321.8103μg/mL,103.7696μg/mL(P㩳0.05);甲基化白藜蘆醇組、乙酰白藜蘆醇組藥物干預(yù)24h和48h后的3T3-L1脂肪細胞分布于Q2象限和Q4象限的比例逐漸增多,細胞凋亡率呈現(xiàn)上升趨勢,表現(xiàn)出較為明顯的時間依賴關(guān)系(P㩳0.05);與空白對照組比較,甲基化白藜蘆醇組、乙酰白藜蘆醇組均下調(diào)3T3-L1脂肪細胞中的PPARγmRNA的表達(P㩳0.05)。 結(jié)論:乙酰白藜蘆醇和甲基化白藜蘆醇均有較好的脂溶性;甲基化白藜蘆醇和乙酰白藜蘆醇對3T3-L1脂肪細胞有抑制作用,可誘導(dǎo)3T3-L1脂肪細胞的凋亡,能下調(diào)3T3-L1脂肪細胞PPARγmRNA的表達水平。
[Abstract]:Objective: to synthesize the derivatives of resveratrol, acetyl resveratrol and methylresveratrol. Their effects on 3 T 3 L 1 adipocytes were observed. Methods: acetyl resveratrol and methylated resveratrol were chemically synthesized from resveratrol. The structures of the products were analyzed by NMR, UV and MS spectra. The cell experiment was divided into blank control group, resveratrol control group, methylated resveratrol group and acetyl resveratrol group. The inhibitory effects of methylated resveratrol and acetyl resveratrol on the proliferation of 3T3-L1 adipocytes were determined by MTT assay, and the effects of methylated resveratrol and acetyl resveratrol on the proliferation of 3T3-L1 adipocytes were observed after 24 h and 48 h treatment, respectively. The apoptosis of 3T3-L1 adipocytes was detected by Annexin V-PI double staining, and the expression of PPAR 緯 mRNA in 3T3-L1 adipocytes was detected by RT-PCR. Results: acetyl resveratrol and methylated resveratrol were synthesized by structure confirmation.
