【摘要】：目的:制備阿德福韋酯納米脂質載體(ADV-NLC),并優(yōu)化其處方。方法:以硬脂酸和單硬脂酸甘油酯為固體脂質,油酸為液體脂質,雙子(Gemini)表面活性劑、聚山梨酯80為乳化劑,十二烷基硫酸鈉(SDS)為穩(wěn)定劑,采用溶劑分散-超聲法制備ADV-NLC。以粒徑、多分散指數(shù)、Zeta電位和包封率為指標,單因素試驗篩選Gemini表面活性劑-聚山梨酯80比例、乳化劑用量(乳化劑與水相之比)、藥脂比、固-液脂質比。結果:處方為乳化劑(雙子表面活性劑-聚山梨酯80比例1∶2)用量3%、藥脂比4.5%,固-液脂質比6∶5。所制ADV-NLC的平均粒徑為(48.83±2.65)nm,多分散指數(shù)0.3,Zeta電位為(-28.7±1.8)m V,包封率為(77.65±0.03)%(n=3)。結論:成功制得ADV-NLC,且處方合理、可行。
[Abstract]:Aim: to prepare adefovir dipivoxil (ADV-NLC) and optimize its formulation. Methods: ADV-NLC was prepared with stearic acid and monostearate as solid lipid, oleic acid as liquid lipid, Gemini as surfactant, polysorbate 80 as emulsifier and sodium dodecyl sulfate as stabilizer. The particle size, polydispersity index, Zeta potential and encapsulation efficiency were used as indexes. The ratio of Gemini surfactant to polysorbate 80, the amount of emulsifier (emulsifier to water phase), the ratio of lipids to lipids, and the ratio of solid to liquid lipid were selected by single factor test. Results: the dosage of emulsifier (1: 2 ratio of Gemini surfactant to polysorbate 80) was 3, the ratio of drug to lipid was 4.5 and the ratio of solid to liquid lipid was 6: 5. The average particle size of ADV-NLC was 48.83 鹵2.65 nm, the polydispersity index was 0.3 鹵1.8mV, and the entrapment efficiency was 77.65 鹵0.03mV. Conclusion: ADV-NLC was successfully prepared and the prescription was reasonable and feasible.
