海洋細菌Halomonas elongate的次級代謝產物研究

【摘要】:目的對具有抗腫瘤活性的海洋細菌Halomonas elongate進行次級代謝產物的分離、純化及抗腫瘤活性研究。方法通過大孔吸附樹脂、硅膠、ODS、Sephadex LH-20凝膠柱色譜等分離純化手段,對H.elongate的次級代謝產物進行分離純化;根據(jù)1H NMR譜、13C NMR譜及MS分析,并結合文獻確定結構;采用MTT法對分離得到的次級代謝產物進行活性檢測。結果從海洋細菌H.elongate的發(fā)酵粗提物中分離鑒定3個化合物,分別為胸腺嘧啶-2'-脫氧核苷(1)、環(huán)(L-甘氨酸-L-脯氨酸)二肽(2)、1-(2'-脫氧-β-D-赤式-呋喃戊糖)-1氫-1,2,4-三嗪酮(3),活性檢測結果顯示3個化合物對人肺癌細胞H1299生長活性均無顯著抑制作用(IC5020μmol/L)。結論化合物1~3均為首次從Halomonas中分離得到。
[Abstract]:Objective to isolate and purify the secondary metabolites of marine bacteria Halomonas elongate with anti-tumor activity and to study its anti-tumor activity. Methods the secondary metabolites of H.elongate were separated and purified by macroporous adsorption resin, Sephadex LH-20 gel column chromatography, 1H NMR spectrum, 13C NMR and MS analysis. The activity of secondary metabolites was determined by MTT method. Results three compounds were isolated and identified from the fermented crude extracts of marine bacteria H.elongate. They were thymidine (Thymine) -2ON-deoxynucleoside (1), cyclic L- glycine (L-proline) dipeptidyl (Thyopyrimidine), dipeptidyl, deoxiodo- 尾-D- (1-triazinone) -2-triazinone (H1299). The results of activity test showed that the three compounds had no significant inhibitory effect on the growth activity of human lung cancer cell line H1299 by IC5020 渭 mol 路L ~ (-1) 路L ~ (-1). Conclusion compounds 1 and 3 were isolated from Halomonas for the first time.
