孕烯醇酮對大鼠空間學習記憶能力和ChAT蛋白表達的影響

【摘要】:目的:研究孕烯醇酮(Pregnenolone,PREG)干預對老年雄性SD大鼠空間學習記憶能力的影響,探討PREG的潛在神經(jīng)保護作用及其機制。方法:24月齡雄性SD大鼠40只,隨機分為對照組(A組)、溶劑對照組(B組)、低劑量PREG(0.5 mg/kg)干預組(C組)、高劑量PREG(2.0 mg/kg)干預組(D組),隔日腹腔注射一個月,利用Y迷宮訓練評估PREG對實驗大鼠空間學習記憶能力的影響,運用免疫熒光技術(shù)和Western Blot檢測分析大鼠記憶相關(guān)腦區(qū)ChAT蛋白表達的變化。結(jié)果:Y迷宮訓練結(jié)果顯示D組大鼠平均連續(xù)正確反應次數(shù)為7.5±1.35,明顯多于其他各組;免疫熒光檢測結(jié)果顯示D組大鼠前額葉、顳葉、海馬內(nèi)ChAT免疫陽性的細胞數(shù)量相應是26.8±2.6、19.8±2.6、25.8±2.4,明顯多于其他處理組;Western Blot檢測顯示D組大鼠前額葉、顳葉、海馬皮層組織ChAT蛋白的表達明顯高于其他各組(P0.05)。結(jié)論:PREG有助于改善老年雄性SD大鼠空間學習記憶能力,其作用機制可能是促進了大鼠記憶相關(guān)腦區(qū)ChAT的表達上調(diào),提高了膽堿能神經(jīng)元細胞的功能活性。
[Abstract]:Aim: to study the effects of Pregnenolone preg on spatial learning and memory in aged male SD rats and to explore the potential neuroprotective effect of PREG and its mechanism. Methods Forty male Sprague-Dawley rats (24 months old) were randomly divided into three groups: control group A, solvent control group B, low dose PREG(0.5 mg / kg group C, high dose PREG(2.0 mg / kg group D, intraperitoneal injection for one month every other day. Y-maze training was used to evaluate the effect of PREG on the spatial learning and memory ability of experimental rats. The changes of ChAT protein expression in memory related brain regions of rats were detected by immunofluorescence and Western Blot. Results the average continuous correct reaction times of group D was 7.5 鹵1.35, which was significantly higher than that of other groups, and the results of immunofluorescence showed that the prefrontal lobe and temporal lobe of D group were significantly higher than those of other groups. The expression of ChAT protein in prefrontal lobe, temporal lobe and hippocampal cortex in group D was significantly higher than that in group D (26.8 鹵2.6 鹵19.8 鹵2.6 鹵25.8 鹵2.4), which was significantly higher than that in other groups. Conclusion: Preg can improve the spatial learning and memory ability of aged male SD rats, and its mechanism may be to promote the up-regulation of ChAT expression in memory related brain regions of rats and to increase the functional activity of cholinergic neurons.
