埃羅替尼對小鼠角膜上皮組織病理學及超微結構的影響

【摘要】:目的探討埃羅替尼對小鼠角膜上皮組織引起的組織病理學及超微結構改變的影響。方法30只雄性6~8周齡BALB/c小鼠,隨機分為對照組(12只)和實驗組(12只),6只不作處理為空白對照。實驗組使用20μmol·L~(-1)埃羅替尼液滴眼,對照組采用PBS滴眼,每天4次。分別在干預后1 d、7 d、14 d對各組小鼠進行熒光素鈉(fluorescent,FL)染色及評分。干預后14 d取小鼠眼球,光學顯微鏡及電子顯微鏡下觀察角膜上皮及細胞的結構變化。提取角膜蛋白進行Western Blot檢測。結果干預前,實驗組和對照組FL評分比較差異無統(tǒng)計學意義(P0.05)。在干預后1 d、7 d、14 d,實驗組FL評分較干預前明顯升高,差異均有統(tǒng)計學意義(均為P0.05),而對照組FL評分與干預前無明顯改變,差異均無統(tǒng)計學意義(均為P0.05);兩組間FL評分在干預后1 d相比,差異無統(tǒng)計學意義(P0.05);兩組間FL評分在干預后7 d、14 d相比,差異均有統(tǒng)計學意義(均為P0.05)。實驗組小鼠角膜上皮細胞排列紊亂,層數(shù)增加;電鏡下見角膜上皮表層細胞形態(tài)不規(guī)則、脫落,微絨毛減少、消失;表層上皮細胞的橋粒、半橋粒連接數(shù)目明顯減少。實驗組角膜表皮生長因子受體表達明顯發(fā)生變化,兩組表皮生長因子受體比較差異有統(tǒng)計學意義(P0.05)。結論埃羅替尼會引起小鼠角膜上皮的組織結構及細胞超微結構損壞。其機制可能是通過抑制表皮生長因子受體的活化來影響角膜上皮的。
[Abstract]:Objective to investigate the effects of erotinib on histopathological and ultrastructural changes induced by corneal epithelium in mice. Methods Thirty male BALB/c mice aged 6 to 8 weeks were randomly divided into control group (n = 12) and experimental group (n = 12). The experimental group was treated with 20 渭 mol L ~ (-1) Erotini drops, while the control group was treated with PBS drops 4 times a day. The mice in each group were stained with sodium fluorescein fluorescein FLL on the 7th day and 14th day after intervention. The cornea epithelium and cell structure were observed by optical microscope and electron microscope 14 days after intervention. Cornea protein was extracted for Western Blot detection. Results before intervention, there was no significant difference in FL score between the experimental group and the control group (P 0.05). The scores of FL in the experimental group were significantly higher than those before the intervention on the 1st day, 7th day and 14th day after the intervention (all P 0.05), but the FL score of the control group had no significant change compared with that before the intervention. There was no significant difference in FL score between the two groups (P 0.05), and there was no significant difference in FL score between the two groups on the 1st day after intervention (P 0.05), and there was significant difference in FL score between the two groups at 7 d and 14 d after intervention (all P 0.05). In the experimental group, the corneal epithelial cells arranged disorderly and the number of layers increased; under the electron microscope, the corneal epithelial surface cells were irregular in shape, shedding off, the microvilli decreased and disappeared; the desmosome and semi-desmosome junctions of the surface epithelial cells decreased significantly. The expression of epidermal growth factor receptor (EGF) in the experimental group was obviously changed, and the difference between the two groups was statistically significant (P 0.05). Conclusion Erotini can damage the tissue structure and cell ultrastructure of corneal epithelium in mice. The mechanism may affect corneal epithelium by inhibiting the activation of epidermal growth factor receptor (EGF).
