高效毛細管電泳法測定大鼠血漿中頭孢地尼濃度及其藥動學研究

【摘要】:目的:測定大鼠血漿中頭孢地尼濃度,并進行藥動學考察。方法:采用高效毛細管電泳法(HPCE)。使用熔融硅膠毛細管柱(75μm,總長為30 cm,有效長度為21.5 cm),緩沖液為20 mmol/L檸檬酸,進樣電壓為10 k V持續(xù)10 s,分離電壓為-20 k V,檢測波長為214 nm。取6只大鼠ig頭孢地尼溶液(20 mg/kg),分別于給藥前及給藥后0.25、0.5、1、1.5、2、3、4、6、12、24 h經(jīng)尾緣靜脈取血0.2 m L進行測定。采用BAPP 2.0軟件計算藥動學參數(shù)。結果:頭孢地尼質(zhì)量濃度在0.2~50μg/m L范圍內(nèi)線性關系良好(r=0.999 7),定量下限為0.2μg/m L;日內(nèi)(n=6)、日間(n=3)精密度試驗的RSD均不大于12.2%;穩(wěn)定性試驗的RSD≤9.10%(n=6);方法回收率為95.4%~114.3%(RSD=9.0%,n=6);基質(zhì)效應為63.5%~70.2%(RSD=10.39%,n=6)。頭孢地尼在大鼠體內(nèi)的t1/2為(0.54±0.01)h,MRT為(1.90±0.14)h,c_(max)為(32.92±0.81)μg/m L,t_(max)為(1.50±0.02)h,AUC_(0-24 h)為(46.65±0.44)μg·h/m L,AUC_(0-∞)為(46.83±0.44)μg·h/m L。結論:該方法快速、準確、簡便,可用于大鼠血漿中頭孢地尼濃度的測定及其藥動學考察。
[Abstract]:Objective: to determine the concentration of cefdinir in rat plasma and to investigate its pharmacokinetics.Methods: high performance capillary electrophoresis (HPCE) was used.The fused silica gel capillary column was used for 75 渭 m, the total length was 30 cm, the effective length was 21.5 cm ~ (-1), the buffer solution was 20 mmol/L citric acid, the injection voltage was 10 kV for 10 s, the separation voltage was -20 kV, the detection wavelength was 214 nm.Before and after administration of cefdinir solution, 0.2 mL of blood was collected from 6 rats before and after administration of cefodienil (0.25g / kg), respectively.
