【摘要】：目的:建立高效、靈敏、快速的超高效液相色譜-電解質(zhì)-質(zhì)譜/質(zhì)譜(UPLC-ESI-MS/MS)測定人血漿中克拉霉素濃度的方法,并應用于藥動學研究。方法:血漿樣品先經(jīng)碳酸鈉溶液堿化,經(jīng)乙醚液-液萃取后,以0.05 mol·L~(-1)乙酸銨水溶液-乙腈為流動相進行梯度洗脫,色譜柱采用BEH C_(18)(50 mm×2.1 mm,1.7μm)色譜柱,克拉霉素和內(nèi)標羅紅霉素定量分析的離子對分別為m/z748.6→157.9和m/z837.0→679.2。結果:克拉霉素的線性范圍為1~3 000 ng·mL~(-1)且線性關系良好,日內(nèi)日間精密度均小于10.0%,準確度在-7.6%~6.0%之間,樣品運行時間為2.0min。結論:該法靈敏、快速,適用于克拉霉素高通量樣本的分析,已成功應用于克拉霉素人體藥動學研究。
[Abstract]:Objective: to establish a high performance, sensitive and rapid method for the determination of clarithromycin in human plasma by UPLC-ESI-MS / MS.Methods: plasma samples were first alkalized by sodium carbonate solution, then extracted by ether liquid-liquid, then gradient elution was carried out with 0.05 mol ammonium acetate aqueous solution and acetonitrile as mobile phase. The chromatographic column was BEH C20 18mm 脳 2.1 mm ~ (1.7 渭 m) column.The ion pairs of clarithromycin and roxithromycin were m/z748.6 157.9 and m/z837.0 679.2, respectively.Results: the linear range of clarithromycin was 1 ~ 3 000 ng / mL 路L ~ (-1) and the linear relationship was good. The intra-day precision of clarithromycin was less than 10.0, the accuracy was between -7.6% and 6.0%, and the sample running time was 2.0 min.Conclusion: this method is sensitive, rapid and suitable for the analysis of clarithromycin in high throughput samples, and has been successfully applied to the pharmacokinetics of clarithromycin in human body.
