【摘要】：目的研究脂氧素對糖尿病大鼠心肌缺血再灌注損傷心肌凋亡細胞及表達的影響。方法按照體重將SD大鼠隨機分為3組:假手術(shù)組、模型組和實驗組。先建立糖尿病大鼠模型,然后實驗組給予脂氧素0.1 mg·kg~(-1),再建立心肌缺血再灌注損傷模型。計算細胞凋亡指數(shù);用免疫印跡法和RT-PCR法檢測心肌組織Bax蛋白、半胱氨酸天冬氨酸蛋白酶3(Caspase-3);用酶聯(lián)免疫吸附法檢測腫瘤壞死因子-α(TNF-α)、白細胞介素~(-1)(IL~(-1))、白細胞介素-6(IL-6)、白細胞介素-8(L-8)含量。結(jié)果假手術(shù)組、模型組與實驗組的心肌細胞凋亡指數(shù)分別為(4.96±0.61)%,(16.72±1.96)%,(8.95±0.99)%,模型組的心肌細胞凋亡指數(shù)高于假手術(shù)組,實驗組的心肌細胞凋亡指數(shù)低于模型組,差異有統(tǒng)計學意義(均P0.05)。模型組大鼠的血清TNF-α為(52.29±6.84)ng·L~(-1)、IL~(-1)為(24.49±3.74)ng·L~(-1)、IL-6為(314.59±45.64)ng·m L~(-1)、IL-8為(223.49±34.59)ng·m L~(-1);均高于假手術(shù)組的TNF-α為(19.62±2.42)ng·L~(-1)、IL~(-1)為(9.18±1.05)ng·L~(-1)、IL-6為(114.52±14.54)ng·m L~(-1)、IL-8為(84.59±10.12)ng·m L~(-1);而實驗組血清TNF-α為(30.39±4.65)ng·L~(-1)、IL~(-1)為(13.52±1.95)ng·L~(-1)、IL-6為(189.46±23.49)ng·m L~(-1)、IL-8為(135.19±17.85)ng·m L~(-1),模型組與假手術(shù)比較或?qū)嶒灲M與模型組比較,差異均有統(tǒng)計學意義(均P0.05)。模型組、假手術(shù)和實驗組大鼠的心肌組織中mRNA Bax表達量分別為249.55±31.23,100.00±14.52,155.49±18.49,而Caspase-3表達量分別為264.95±34.52,100±16.09,170.19±19.52;Bax蛋白表達量分別為221.38±19.45,100.00±15.68,135.54±16.79;Caspase-3蛋白表達量分別為305.69±35.62,100.00±16.10,145.62±21.34。模型組高于假手術(shù)組、而實驗組低于模型組,組間比較差異均有統(tǒng)計學意義(均P0.05)。結(jié)論脂氧素對糖尿病大鼠心肌缺血再灌注損傷具有保護作用,能夠減少細胞凋亡數(shù)目并抑制凋亡基因與蛋白表達。
[Abstract]:Objective to study the effect of lipoxygenin on myocardial apoptotic cells and expression of myocardial ischemia-reperfusion injury in diabetic rats. Methods SD rats were randomly divided into three groups according to their body weight: sham operation group, model group and experimental group. Then the experimental group was given lipoxygenin 0.1 mg / kg to establish myocardial ischemia-reperfusion injury model. Apoptosis index was calculated, and Bax protein in myocardial tissue was detected by Western blotting and RT-PCR assay. Cysteine aspartate proteinase 3 (caspase-3) was used to detect the contents of tumor necrosis factor- 偽 (TNF- 偽), interleukin-1 (IL-1), interleukin-6 (IL-6), interleukin-8 (IL-8) by enzyme-linked immunosorbent assay (Elisa). The myocardial apoptosis index of the model group and the experimental group were 4.96 鹵0.61g and 16.72 鹵1.96 respectively. The myocardial apoptosis index of the model group was higher than that of the sham operation group, and the myocardial apoptosis index of the experimental group was lower than that of the model group. There was significant difference in serum TNF- 偽 between model group and sham operation group (P 0.05) (all P0.050.The serum TNF- 偽 of model group was 19.62 鹵6.84)ng Ln-1C ~ (-1)): IL-6 was 24.49 鹵3.74)ng L ~ (-1), IL-6 was 314.59 鹵45.64)ng ~ (-1) 45.64)ng ~ (-1) IL-8 was 223.49 鹵34.59)ng ~ (-1), which was higher than that of sham-operated group ((19.62 鹵2.42)ng) ~ (-1)). IL-6 was (114.52 鹵14.54)ng) -1L ~ (-1) and (84.59 鹵10.12)ng ~ (1)) in the experimental group was (30.39 鹵4.65)ng) -11L ~ (-1), and that in the experimental group was (30.39 鹵4.65)ng) -11L ~ (-1), and that in the experimental group was significantly higher than that in the control group (P < 0.01), and that in the experimental group was significantly higher than that in the sham operation group (9.62 鹵1.05)ng / L ~ (-1)), and that in the experimental group was (114.52 鹵14.54)ng) m ~ (-1) and (84.59 鹵10.12)ng ~ (-1)) respectively, respectively, and that in the experimental group was (30.39 鹵4.65)ng) -11L ~ (-1), and that in the experimental group was significantly higher than that in the sham operation group (n = 19.62 鹵2.42)ng), and that in the experimental group was higher than that in the sham operation group (n = 9.18 鹵1.05)ng), and that in the experimental group was (114.52 鹵14.54)ng) -1L ~ (-1). Sham-operation comparison or comparison between experimental group and model group, The expression of mRNA Bax in model group, sham operation group and experimental group were 249.55 鹵31.23100.00 鹵14.52155.49 鹵18.49, and Caspase-3 expression levels were 264.95 鹵34.52100 鹵16.09170.19 鹵19.52Bax protein respectively (221.38 鹵19.45100.00 鹵15.68135.54 鹵16.79caspase-3). The expression of Caspase-3 protein in model group was higher than that in sham operation group (305.69 鹵35.2100.00 鹵16.10145.62 鹵21.34. respectively). But the experimental group was lower than the model group, and the difference between the two groups was statistically significant (P 0.05). Conclusion lipoxygenin has protective effect on myocardial ischemia-reperfusion injury in diabetic rats, can reduce the number of apoptosis and inhibit the expression of apoptotic gene and protein.
