微波消解-電感耦合等離子體質(zhì)譜法測(cè)定大鼠全血中納米銀

【摘要】:目的:建立和確證一種簡(jiǎn)便、準(zhǔn)確的測(cè)定大鼠全血中銀濃度的分析方法。方法:選取銦(115In)為內(nèi)標(biāo),先用硝酸對(duì)全血樣品進(jìn)行微波消解處理,再采用電感耦合等離子體質(zhì)譜法測(cè)定消化液中銀離子的濃度。結(jié)果:內(nèi)源性物質(zhì)不干擾銀和內(nèi)標(biāo)的測(cè)定,全血中銀的濃度在5.0~1 000μg·L-1范圍內(nèi)線性關(guān)系良好,日內(nèi)精密度RSD≤3.3%,日間精密度RSD≤3.8%;提取回收率為97.5%~101.9%,基質(zhì)效應(yīng)為97.4%~99.4%。應(yīng)用該法研究了大鼠靜脈注射納米銀后銀的體內(nèi)經(jīng)時(shí)變化,結(jié)果表明,給藥(40 nm)后,大鼠血液中銀濃度先迅速下降而后上升,之后再緩慢消除。結(jié)論:該方法可為納米銀在生物機(jī)體內(nèi)的評(píng)價(jià)研究提供分析方法參考。
[Abstract]:Objective: to establish and confirm a simple and accurate method for the determination of silver in whole blood of rats. Inductively coupled plasma mass spectrometry (ICP-MS) was used to determine the concentration of silver ion in digestive fluid.
