【摘要】：目的探討不同血小板制劑對(duì)細(xì)胞體外增殖活力的影響,并對(duì)其作用機(jī)制進(jìn)行初步研究。方法制備不同血小板制劑:新鮮機(jī)采血小板、冰凍機(jī)采血小板、富血小板凝膠上清、乏血小板血漿等,以1640培養(yǎng)基為對(duì)照,與MM.1S細(xì)胞(多發(fā)性骨髓瘤細(xì)胞)在相同的培養(yǎng)條件下進(jìn)行共培養(yǎng),用流式細(xì)胞儀對(duì)培養(yǎng)后的細(xì)胞進(jìn)行細(xì)胞計(jì)數(shù),觀察細(xì)胞的增殖情況。結(jié)果 1)單獨(dú)的血小板及乏血小板血漿均對(duì)MM.1S細(xì)胞有促增殖作用,且促增殖效果與樣品濃度成正相關(guān)。2)新鮮機(jī)采血小板對(duì)MM.1S細(xì)胞促增殖作用在各實(shí)驗(yàn)組中最高,新鮮機(jī)采凝膠上清和乏血小板血漿次之;二甲基亞砜對(duì)細(xì)胞增殖有抑制作用;凝血酶對(duì)細(xì)胞增殖無明顯影響。3)血小板經(jīng)過冰凍處理后,機(jī)采冰凍血小板與冰凍凝膠上清對(duì)MM.1S細(xì)胞促增殖作用較新鮮機(jī)采血小板和凝膠上清減弱。但機(jī)采冰凍血小板與冰凍凝膠上清對(duì)MM.1S細(xì)胞的促增殖作用存在劑量效應(yīng),在稀釋濃度40-80倍左右時(shí),具有相對(duì)較強(qiáng)的促增殖影響力。結(jié)論不同血小板制劑對(duì)MM.1S細(xì)胞的促增殖作用各有不同,此研究將為制備血小板凝膠選擇原輔材料提供實(shí)驗(yàn)依據(jù)和參考。
[Abstract]:Objective to investigate the effect of different platelet preparations on cell proliferation in vitro and its mechanism. Methods different platelets were prepared: fresh platelets, cryopreserved platelets, platelets supernatants, and platelet-rich gel supernatants. The platelet-deficient plasma was co-cultured with MM.1S cells (multiple myeloma cells) in 1640 medium, and the cells were counted by flow cytometry. Results 1) platelets and platelets alone could promote the proliferation of MM.1S cells. The effect of promoting proliferation was positively correlated with the concentration of sample. 2) the effect of fresh platelets on the proliferation of MM.1S cells was the highest, the supernatant of fresh mechanized gel and the plasma of lack of platelets were the second, and dimethyl sulfoxide inhibited the proliferation of MM.1S cells. Thrombin has no significant effect on cell proliferation. The effect of frozen platelets and supernatants on the proliferation of MM.1S cells was weaker than that of fresh platelets and supernatants of gel, but there was a dose effect on the proliferation of MM.1S cells by the supernatant of frozen platelets and supernatants of frozen gels. Conclusion the effects of different platelet preparations on the proliferation of MM.1S cells are different. This study will provide experimental basis and reference for the preparation of raw and auxiliary materials for platelet-gel selection.
