阿霉素對腎上腺皮質癌細胞系SW-13克隆特性的影響

【摘要】:目的:研究阿霉素在體干預對腎上腺皮質癌系SW-13克隆特性的影響。方法:通過建立阿霉素干預裸鼠連續(xù)傳代SW-13細胞系皮下移植瘤模型,將空白組及實驗組細胞進行單細胞克隆行成試驗,觀察單個細胞生長曲線,并計算克隆形成率及3種克隆(全克隆、部分克隆和旁克隆)的百分比,并進行Hoechst33342試驗檢測細胞的耐藥性。結果:SW-13細胞系經過阿霉素干預裸鼠連續(xù)傳代移植瘤模型后,克隆形成率較空白對照組增加(P0.05);全克隆的百分比高于空白對照組(P0.05);經Hoechst33342試驗發(fā)現經阿霉素干預腎上腺皮質癌細胞熒光減弱。結論:阿霉素在體干預提高腎上腺皮質癌細胞系SW-13的克隆形成能力及全克隆形成能力,并提高其耐藥性。
[Abstract]:Objective: To study the effect of adriamycin intervention SW-13 clone characteristics of adrenal cortical carcinoma lines in vivo. Methods: through the establishment of adriamycin intervention continuous passaging in nude mice SW-13 cells subcutaneous transplantation tumor model, the blank group and the experimental group were single cell cloned into a single test, observe the cell growth curve and colony forming efficiency was calculated and 3 clone (Quan Kelong, part of cloning and cloning by) the percentage of drug resistance and Hoechst33342 test cells. Results: SW-13 cells after adriamycin intervention after continuous passaging in nude mice xenograft model, clone formation rate than the control group increased (P0.05); the percentage of total clones higher than the control group (P0.05); the the Hoechst33342 trial found that after adriamycin treatment of adrenal cortical carcinoma cells. Conclusion: the fluorescence decrease of adriamycin in body intervention to improve the adrenocortical carcinoma cell line cloned SW-13 formation Force and total cloning ability and increase its resistance.
