維生素C對(duì)青春期鄰苯二甲酸二丁酯暴露致大鼠卵巢氧化應(yīng)激的干預(yù)作用

【摘要】:目的探討維生素C(vitamin C,Vit C)對(duì)青春期鄰苯二甲酸二丁酯(dibutyl phthalate,DBP)暴露致大鼠卵巢氧化應(yīng)激的干預(yù)作用。方法將160只健康初斷乳SPF級(jí)Wistar雌性大鼠隨機(jī)分為5組,分別為對(duì)照(玉米油)組和低(100mg/kg)、高(500 mg/kg)劑量DBP暴露組及低(100 mg/kg)、高(500 mg/kg)劑量DBP+Vit C(125 g/L)干預(yù)組。采用灌胃方式暴露DBP,暴露容量為10 ml/kg,每天1次;采用自由飲水方式暴露Vit C,連續(xù)暴露30 d。分別于暴露第5、10、20、30天,測(cè)定大鼠卵巢組織中MDA、GSH含量和SOD、CAT、GSH-Px活力。結(jié)果與對(duì)照組相比,DBP暴露組大鼠卵巢組織中MDA含量均升高,而GSH含量和SOD、CAT、GSH-Px活力均下降;與相同劑量DBP暴露組相比,Vit C干預(yù)組大鼠卵巢組織中MDA均下降,而GSH含量和SOD、CAT、GSH-Px活力均升高。且隨著DBP暴露劑量的升高,DBP暴露組和Vit C干預(yù)組大鼠卵巢組織中的GSH含量和SOD、CAT、GSH-Px活力均呈下降趨勢(shì),而MDA含量呈上升趨勢(shì)。結(jié)論 DBP暴露可致青春期雌性大鼠卵巢組織產(chǎn)生脂質(zhì)過(guò)氧化反應(yīng),誘導(dǎo)氧化應(yīng)激,導(dǎo)致卵巢抗氧化能力下降;而抗氧化劑Vit C對(duì)于DBP所致生殖系統(tǒng)的氧化損傷具有一定的拮抗作用。
[Abstract]:Objective to investigate the effect of vitamin C vitamin C (Vit C) on oxidative stress induced by dibutyl phthalate dibutyl phthalate (DBP) in pubertal rats. Methods 160 healthy SPF female rats were randomly divided into 5 groups. They were the control group (corn oil), the low dose of DBP (100mg / kg), the low dose of DBP (100mg / kg) and the group of low dose (100mg / kg) DBP Vit (125g / L). The exposure capacity was 10ml / kg, once a day, and the dose of DBP Vit (125g / L) was 100 mg / kg, 100 mg / kg, 100 mg / kg, 100 mg / kg, 100 mg / kg, 100 mg / kg, 100 mg / kg, 100 mg / kg, 100 mg / kg respectively, respectively. Vit C was exposed to free drinking water for 30 days. The content of MDA-GSH and the activity of MDA in ovarian tissue of rats were measured on the 20th day after exposure. Results compared with the control group, the content of MDA in the ovarian tissue of the rats exposed to Vit was higher than that of the control group. The content of GSH and the activity of GSH-Px in DBP were decreased, and the MDA in ovarian tissue of Vit C group was decreased compared with the same dose of DBP exposure group. However, the content of GSH and the activity of GSH-Px in DBP increased, and the content of GSH and the activity of GSH-Px in the ovary of rats exposed to DBP and treated with Vit C showed a decreasing trend with the increase of DBP exposure dose. Conclusion DBP exposure can induce lipid peroxidation in the ovary of adolescent female rats, induce oxidative stress and decrease the antioxidant capacity of ovary. The antioxidant Vit C has a certain antagonistic effect on the oxidative damage of reproductive system induced by DBP.
