大鼠血漿中枸杞多糖熒光標記物定量分析方法的建立

【摘要】:建立大鼠血漿中異硫氰酸熒光素標記的枸杞多糖(fluorescein isothiocyanate labeled Lycium barbarumpolysaccharides,LBP-FITC)的熒光定量分析方法,線性方程為y=1 378.7x-14.98(R2=0.998 9),血漿中LBP-FITC質(zhì)量濃度在0.05~10μg/m L范圍內(nèi)與熒光強度呈良好的線性關系。方法的精密度、回收率以及穩(wěn)定性考察分析證明該方法符合生物樣品體內(nèi)定量分析的要求。單次灌胃給予大鼠LBP-FITC后,以藥代動力學Win Nonlin軟件采用非房室模型擬合大鼠單次口服LBP-FITC的經(jīng)時血藥質(zhì)量濃度數(shù)據(jù),計算主要的藥代動力學參數(shù)為最大峰質(zhì)量濃度(Cmax)(2.39±0.71)μg/m L、達峰時間(tmax)(1.17±0.41)h;藥時曲線下面積AUC0-t為(45.85±7.12)(μg?h)/m L,AUC0-∞為(127.31±39.00)(μg?h)/m L;半衰期(t1/2)為(80.32±32.70)h;清除率為(1 693.96±492.47)m L/kg;平均滯留時間為(20.64±1.36)h。
[Abstract]:A method for quantitative analysis of fluorescein isothiocyanate labeled Lycium barumPolysaccharide LBP-FITCin rat plasma with fluorescein isothiocyanate was established. The linear equation is YY 1 378.7x-14.98 9 渭 g / mL LBP-FITC concentration in plasma is linear with fluorescence intensity in the range of 0.05 ~ 10 渭 g / m ~ (-1). The precision of the method is obtained. The results of recovery and stability analysis showed that the method met the requirements of quantitative analysis of biological samples in vivo.
