【摘要】：目的觀察大鼠慢性炎性痛時(shí)背根神經(jīng)節(jié)(dorsal root ganglion,DRG)μ型阿片受體(mu opioid receptor,MOR)表達(dá)的變化及炎癥局部注射MOR激動(dòng)劑和拮抗劑對(duì)痛閾的干預(yù)作用,探討MOR在慢性炎性疼痛中的作用。方法足底注射弗氏完全佐劑(complete freund’s adjuvant,CFA)制備大鼠慢性炎性痛模型,采用免疫組化法檢測(cè)DRG MOR陽(yáng)性細(xì)胞的表達(dá);經(jīng)大鼠足跖背部分別注射MOR激動(dòng)劑、拮抗劑,采用輻射熱法檢測(cè)給藥前后大鼠痛閾的變化。結(jié)果足底注射CFA誘導(dǎo)出大鼠慢性炎性痛,在造模后第18天,痛閾依然低于正常對(duì)照組;免疫組化結(jié)果表明,與正常對(duì)照組相比,CFA大鼠DRG MOR陽(yáng)性細(xì)胞表達(dá)增多(P0.01);足跖背部注射MOR激動(dòng)劑減輕CFA大鼠的疼痛,對(duì)正常大鼠痛閾無(wú)影響;足跖背部注射MOR拮抗劑加重CFA大鼠的疼痛,對(duì)正常大鼠痛閾無(wú)影響。結(jié)論 DRG神經(jīng)元MOR在慢性炎性痛時(shí)表達(dá)上調(diào),參與慢性炎性痛的外周調(diào)控,可能有助于防止慢性痛的進(jìn)一步加重。
[Abstract]:Objective to observe the expression of 渭 type opioid receptor mu opioid receptor moor in dorsal root ganglion (DRG) and the effect of local injection of MOR agonists and antagonists on pain threshold during chronic inflammatory pain in rats. To investigate the role of MOR in chronic inflammatory pain. Methods the rat model of chronic inflammatory pain was established by injection of complete Freund's adjuvant freund's adjuvant CFA. the expression of DRG MOR positive cells was detected by immunohistochemical method, and the MOR agonist was injected into the back of the foot and plantar of rats. Results chronic inflammatory pain was induced by plantar injection of CFA in rats, and the pain threshold was still lower than that in normal control group on the 18th day after the model was made. Compared with the normal control group, the expression of DRG MOR positive cells was increased in CFA rats, MOR agonist was injected into the back of the plantar to relieve the pain of CFA rats, and the pain threshold was not affected in the normal rats. The pain of CFA rats was aggravated by the injection of MOR antagonists on the back of the plantar. Conclusion the expression of MOR in DRG neurons is upregulated during chronic inflammatory pain, which may contribute to the prevention of further exacerbation of chronic pain by participating in the peripheral regulation of chronic inflammatory pain.
