【摘要】：目的觀察羅紅霉素對小鼠氣管環(huán)的舒張作用,并探討其作用機制。方法取BALB/c小鼠15只,處死后分離氣管環(huán),將氣管環(huán)置于灌流槽中,經(jīng)過高鈣液平衡后開始實驗。使用激動劑乙酰膽堿(Ach)100μmol/L收縮氣管環(huán),待氣管環(huán)張力達到平臺期后,分別加入終濃度為10~(-2)、10~(-1.5)、10~(-1)、10~(-0.5)、10~0、10~(0.5)、10~(0.75)、10~1mg/mL的羅紅霉素,觀察不同濃度羅紅霉素作用下的氣管環(huán)肌張力和舒張比率,將引起氣管環(huán)最大舒張的最低藥物濃度10~(0.75)mg/mL作為羅紅霉素最佳藥物濃度。向灌流槽中加入100μmol/L Ach預收縮氣管環(huán),隨后加入L型鈣離子通道抑制劑硝苯地平、非選擇性陽離子通道抑制劑吡唑鹽3(Pyr3),待抑制劑舒張氣管作用達到平臺期后,加入10~(0.75)mg/mL羅紅霉素,觀察氣管環(huán)的肌張力變化和舒張比率。結(jié)果 Ach預收縮的氣管環(huán)的肌張力為(7.03±0.35)mN,加入最佳濃度的羅紅霉素后,肌張力降至(3.47±0.29)mN(P0.05)。加入硝苯地平、Pyr3后氣管環(huán)的肌張力降低,舒張比率增加(P均0.05);繼續(xù)加入羅紅霉素后,氣管環(huán)的肌張力繼續(xù)降低,舒張比率較前升高(P均0.05)。結(jié)論羅紅霉素對小鼠Ach預收縮的氣管環(huán)具有舒張作用,并可增強硝苯地平、Pyr3對于Ach預收縮氣管環(huán)的舒張作用,其主要通過調(diào)控鈣離子通道來發(fā)揮作用。
[Abstract]:Objective to observe the relaxation effect of roxithromycin on tracheal rings in mice and to explore its mechanism. After the high calcium solution balance, the experiment began. The agonist acetylcholine acetylcholine acetylcholine #number0# 渭 mol/L was used to contract the trachea ring. After the tension of the trachea ring reached the plateau stage, the final concentration of roxithromycin was 10 ~ (10) ~ 10 ~ (-1) ~ (10 ~ (10)) ~ (10 ~ (10)) ~ (10 ~ (10)) (n = 10). To observe the tension and relaxation ratio of tracheal ring muscle under different concentrations of roxithromycin, the lowest drug concentration (10 ~ 0.75 mg / mL) was used as the best concentration of roxithromycin, and the 100 渭 mol/L Ach precontracted tracheal ring was added to the perfusion tank. The L-type calcium channel inhibitor nifedipine and the nonselective cationic channel inhibitor pyrazolium 3 were added. After the relaxation of trachea reached the plateau stage, the inhibitor was added with roxithromycin 0.75 mg / mL. Results the muscle tension of tracheal rings precontracted by Ach was 7.03 鹵0.35 mN.The muscle tension of tracheal rings decreased to 3.47 鹵0.29 mNP0.05 after adding roxithromycin at the optimum concentration, and the muscle tension of tracheal rings decreased after adding nifedipine to Pyr3. After adding roxithromycin, the muscle tension of tracheal rings continued to decrease and the relaxation ratio increased to 0.05%. Conclusion roxithromycin has a relaxing effect on Ach precontracted tracheal rings in mice. The relaxation effect of nifedipine PYR3 on Ach precontracted tracheal rings was enhanced by regulating calcium channels.
