當(dāng)歸小分子多糖對(duì)血管性癡呆大鼠學(xué)習(xí)與記憶功能的影響

【摘要】:目的觀察當(dāng)歸小分子多糖對(duì)血管性癡呆模型大鼠學(xué)習(xí)與記憶功能的影響。方法動(dòng)物隨機(jī)分為6組:假手術(shù)組、模型組、對(duì)照組;3個(gè)劑量當(dāng)歸小分子多糖組。用頸內(nèi)動(dòng)脈注射血栓的方法制作血管性癡呆大鼠模型,當(dāng)歸小分子多糖低、中、高3個(gè)劑量組分別腹腔注射當(dāng)歸小分子多糖0.08,0.15,0.30 g·kg-1,對(duì)照組腹腔注射血栓通注射液42 mg·kg-1,模型組和假手術(shù)組動(dòng)物分別腹腔注射等量0.9%氯化鈉,每天1次,共28 d。用Morris水迷宮實(shí)驗(yàn)觀察當(dāng)歸小分子多糖對(duì)實(shí)驗(yàn)動(dòng)物學(xué)習(xí)及記憶功能的影響,并觀察大鼠腦Tau蛋白的表達(dá)。結(jié)果當(dāng)歸小分子多糖能明顯縮短大鼠的逃避潛伏期,增加停留原平臺(tái)位置時(shí)間;當(dāng)歸小分子多糖中、大2個(gè)劑量組大鼠腦總Tau蛋白表達(dá)明顯下降。結(jié)論當(dāng)歸小分子多糖可改善血管性癡呆大鼠的學(xué)習(xí)及記憶功能,其機(jī)制可能與抑制Tau蛋白表達(dá)的有關(guān)。
[Abstract]:Objective to observe the effect of Angelica sinensis small molecule polysaccharide on learning and memory in vascular dementia rats. Methods the rats were randomly divided into six groups: sham operation group and model group. Control group; three doses of Angelica sinensis small molecule polysaccharide group. Vascular dementia rat model was made by internal carotid artery injection of thrombus. The rats in the high dose group were given intraperitoneal injection of Angelica sinensis small molecular polysaccharide 0.08 ~ 0.15g 路kg ~ (-1), the control group was given 42 mg 路kg ~ (-1) Xue Tong injection, and the model group and the sham operation group were respectively injected with the same amount of 0.9% sodium chloride once a day. The effects of Danggui small molecule polysaccharide on learning and memory function and the expression of Tau protein in rat brain were observed by Morris water maze test. Results the escape latency of rats was significantly shortened by Danggui small molecule polysaccharide. The expression of total Tau protein in rat brain was significantly decreased in the two dose groups. Conclusion Danggui small molecular polysaccharide can improve the learning and memory function of vascular dementia rats. The mechanism may be related to the inhibition of Tau protein expression.
