【摘要】：目的:對注射用達托霉素有關(guān)物質(zhì)測定方法進行優(yōu)化。方法:采用IB-SIL C-8(250 mm×4.6 mm,5μm)色譜柱;以0.039 mol·L~(-1)磷酸二氫銨(用1 mol·L~(-1)磷酸溶液調(diào)節(jié)pH至3.25)-乙腈(80∶20)為流動相A,以0.039 mol·L~(-1)磷酸二氫銨(用1 mol·L~(-1)磷酸溶液調(diào)節(jié)pH至3.25)-乙腈(50∶50)為流動相B;柱溫:20℃;流速:1.5 ml·min~(-1);檢測波長:214 nm。結(jié)果:注射用達托霉素在0.39~77.25#g·ml~(-1)線性范圍內(nèi)濃度與峰面積呈良好線性關(guān)系,r=1.000 0,最低檢測限為2.4 ng,達托霉素與已知和未知雜質(zhì)分離度良好。結(jié)論:本法可用于注射用達托霉素的有關(guān)物質(zhì)的測定。
