【摘要】：目的:采用液相色譜-質(zhì)譜串聯(lián)技術(shù)(HPLC-MS/MS),建立測(cè)定小鼠肺組織中美羅培南含量的方法,為研究美羅培南在肺內(nèi)分布的藥動(dòng)學(xué)奠定基礎(chǔ)。方法:采用Diamonsil C18(150 mm×4.6 mm,5μm)色譜柱進(jìn)行分離,流動(dòng)相為乙腈-0.1%甲酸水,梯度洗脫,流速1.0mL·min~(-1),內(nèi)標(biāo)選擇厄他培南。電噴霧離子化源(ESI),正負(fù)離子切換,美羅培南為正離子檢測(cè)模式,內(nèi)標(biāo)厄他培南為負(fù)離子檢測(cè)模式。多反應(yīng)監(jiān)測(cè)(MRM)方式進(jìn)行定量分析,用于定量分析的離子對(duì)分別為美羅培南m/z384.1→141.2,內(nèi)標(biāo)厄他培南m/z474.4→265.2。結(jié)果:小鼠肺組織勻漿中美羅培南在15.625~2 000 ng·mL~(-1)范圍內(nèi)線性關(guān)系良好(r=0.999 7),平均提取回收率為98.05%~102.54%,日內(nèi)、日間精密度均小于15%。結(jié)論:采用本法測(cè)定小鼠肺組織中美羅培南的濃度,靈敏度高,專屬性強(qiáng),可用于肺組織樣品中美羅培南快速有效的分析以及美羅培南在肺組織分布的藥動(dòng)學(xué)研究。
[Abstract]:Objective: to establish a method for the determination of meropenem in lung tissue of mice by HPLC MS / MS / MS, and to establish a method for the study of the pharmacokinetics of meropenem in the lung. Methods: the chromatographic column of Diamonsil C18C 150mm 脳 4.6mm ~ 5 渭 m was used to separate meropenem from mice. The mobile phase consisted of acetonitrile-0.1% formic acid water, gradient elution, flow rate of 1.0 mL 路min ~ (-1) ~ (-1), internal standard selection of ertapenem, electrospray ionization source (ESI), positive and negative ion switching, meropenem as positive ion detection mode. The internal standard of ertapenem was negative ion detection mode.
