【摘要】：目的:研究健康受試者空腹及高脂高熱量飲食情況下口服硫酸氫氯吡格雷片的藥動學(xué)特征。方法:60名男性健康志愿者單劑量、自身交叉口服硫酸氫氯吡格雷片75 mg。采用液相色譜-質(zhì)譜聯(lián)用(HPLC-MS/MS)法測定人血漿中氯吡格雷的濃度。用DAS3.2.3藥動學(xué)軟件計算藥動學(xué)參數(shù),并用SPSS 17.0軟件對主要參數(shù)進(jìn)行統(tǒng)計分析。結(jié)果:空腹與進(jìn)食后的主要藥動學(xué)參數(shù)如下:Cmax分別為(1 440±2 397)ng·L-1和(4 155±2 117)ng·L-1,AUC0-36分別為(2 268±3 887)ng·L-1·h和(8 691±3 628)ng·L-1·h,AUC0-∞分別為(2 324±3 899)ng·L-1·h和(8 816±3 668)ng·L-1·h,t1/2分別為(5.7±4.7)h和(8.8±3.8)h,tmax分別為(0.7±0.5)h和(1.7±0.7)h,Vd分別為(520 115±471 187)L和(118 826±59 077)L,CL分別為(82 365±70 072)L·h-1和(9 949±4 017)L·h-1,MRT0-36分別為(3.0±1.8)h和(3.6±0.9)h。結(jié)論:高脂高熱量飲食對硫酸氫氯吡格雷片的藥動學(xué)特征有顯著影響,氯吡格雷的達(dá)峰時間延長,生物利用度提高,半衰期延長。
[Abstract]:Objective: to study the pharmacokinetic characteristics of oral clopidogrel sulfate tablets in healthy volunteers on an empty stomach and high fat and high calorie diet. The concentration of clopidogrel in human plasma was determined by liquid chromatography-mass spectrometry (LC-MS / MS). The pharmacokinetic parameters were calculated by DAS3.2.3 pharmacokinetic software. The main pharmacokinetic parameters were analyzed by SPSS 17.0 software.
