氟西汀對慢性應(yīng)激大鼠前額葉谷氨酸轉(zhuǎn)運體GLT-1表達的影響

【摘要】:目的研究氟西汀對慢性應(yīng)激大鼠前額葉谷氨酸轉(zhuǎn)運體1(GLT-1)的影響,進一步探討氟西汀抗抑郁作用可能的分子機制。方法正常SD大鼠60只,隨機分為對照組、慢性不可預(yù)見性應(yīng)激(CUS)組和氟西汀組。對CUS組和氟西汀組大鼠進行CUS應(yīng)激后,氟西汀組給予氟西汀治療,對照組和CUS組給予生理鹽水。實驗結(jié)束后進行糖水偏好和曠場行為測試,并使用免疫組織化學(xué)法和蛋白印跡分析檢測大鼠前額葉GLT-1的表達水平。結(jié)果 (1)行為學(xué)測試結(jié)果顯示,CUS組大鼠糖水偏好、總行程、平均移動速度及直立次數(shù)均低于對照組(P0.01);氟西汀組上述指標均高于CUS組(P0.01)。(2)免疫組織化學(xué)法分析顯示,CUS組與對照組比較,大鼠前額葉GLT-1表達下降(P0.01);經(jīng)氟西汀治療后,大鼠前額葉GLT-1表達比CUS組明顯升高(P0.01)。(3)蛋白印跡分析顯示,CUS組與對照組比較,大鼠前額葉GLT-1表達下降(P0.01);經(jīng)氟西汀治療后,大鼠前額葉GLT-1表達比CUS明顯升高(P0.01)。結(jié)論慢性應(yīng)激下調(diào)大鼠前額葉GLT-1表達水平,而氟西汀上調(diào)GLT-1表達水平,GLT-1表達增加可能是氟西汀抗抑郁作用的分子機制之一。
[Abstract]:Objective to study the effect of fluoxetine on the prefrontal glutamate transporter (GLT-1) in rats under chronic stress and to explore the possible molecular mechanism of antidepressant effect of fluoxetine. The rats in CUS group and fluoxetine group were treated with fluoxetine after CUS stress. The control group and CUS group were given normal saline. After the experiment, sugar water preference and open-field behavior were tested. Immunohistochemical method and Western blot analysis were used to detect the expression of GLT-1 in prefrontal lobe of rats. The average moving speed and the number of upright movement were lower than that of the control group (P 0.01). The above indexes in fluoxetine group were higher than those in CUS group (P 0.01). The immunohistochemical analysis showed that the CUS group was compared with the control group. The expression of GLT-1 in prefrontal lobe of rats was decreased (P 0.01). After fluoxetine treatment, the expression of GLT-1 in the prefrontal lobe of rats was significantly higher than that in the CUS group. Western blot analysis showed that the expression of GLT-1 in the prefrontal lobe of rats was higher than that in the control group. The expression of GLT-1 in prefrontal lobe of rats was decreased (P 0.01). The expression of GLT-1 in prefrontal lobe of rats was significantly higher than that of CUS after fluoxetine treatment. Conclusion chronic stress can down-regulate the expression of GLT-1 in prefrontal lobe of rats. But fluoxetine upregulated GLT-1 expression and GLT-1 expression may be one of the molecular mechanisms of fluoxetine antidepressant effect.
